Mosaicism in health and disease - clones picking up speed.

Post-zygotic variation refers to genetic changes that arise in the soma of an individual and that are not usually inherited by the next generation. Although there is a paucity of research on such variation, emerging studies show that it is common: individuals are complex mosaics of genetically distinct cells, to such an extent that no two somatic cells are likely to have the exact same genome. Although most types of mutation can be involved in post-zygotic variation, structural genetic variants are likely to leave the largest genomic footprint. Somatic variation has diverse physiological roles and pathological consequences, particularly when acquired variants influence the clonal trajectories of the affected cells. Post-zygotic variation is an important confounder in medical genetic testing and a promising avenue for research: future studies could involve analyses of sorted and single cells from multiple tissue types to fully explore its potential.

Genomic and bioacoustic variation in a midwife toad hybrid zone: A role for reinforcement?

PLoS One

November 2024

Laboratory for Amphibian Systematics and Evolution, College of Biology & the Environment, Nanjing Forestry University, Nanjing, China.

Article Synopsis
  • Hybrid zones are areas where diverging species meet, mix their genes, and provide insights into how species diverge and become reproductively isolated.
  • This study focuses on a hybrid zone between two species of midwife toads in southern France, using advanced techniques to analyze genetics and mating calls, aiming to understand genetic mixing and potential reinforcement of reproductive barriers.
  • Findings indicate a narrow hybrid zone with limited gene flow and evidence of reproductive isolation, suggesting that while mating calls overlap less in the hybrid zone, this trend does not lead to clear differences in calls, indicating complex dynamics in reinforcement processes.

Whole genome sequencing study of identical twins discordant for psychosis.

Transl Psychiatry

July 2024

Neuropsychiatric Genetics Research Group, Department of Psychiatry, Trinity College Dublin, Dublin, Ireland.

Article Synopsis
  • Monozygotic (MZ) twins, typically considered genetically identical, actually can have differing DNA variants due to early post-zygotic events, which may relate to differences in disease manifestation.
  • In a study involving whole genome sequencing of 17 pairs of MZ twins discordant for psychotic disorders, researchers found rare genetic variants unique to affected individuals, including specific genes linked to schizophrenia and bipolar disorder.
  • The research identified significant genetic variations, such as deleterious missense variants and rare genic copy number variants (CNVs), suggesting a potential link to the disorders, and emphasizes the need for further investigation in this area.

Application of multiple mosaic callers improves post-zygotic mutation detection from exome sequencing data.

Purpose: The gold standard for identification of post-zygotic variants (PZVs) is droplet digital polymerase chain reaction or high-depth sequencing across multiple tissues types. These approaches are yet to be systematically implemented for monogenic disorders. We developed PZV detection pipelines for correct classification of de novo variants.

Understanding the mechanisms of polyploidization in cardiomyocytes is crucial for advancing strategies to stimulate myocardial regeneration. Although endoreplication has long been considered the primary source of polyploid human cardiomyocytes, recent animal work suggests the potential for cardiomyocyte fusion. Moreover, the effects of polyploidization on the genomic-transcriptomic repertoire of human cardiomyocytes have not been studied previously.

Chromosomal rearrangements are often associated with playing a role in the speciation process. However, the underlying mechanism that favors the genetic isolation associated with chromosomal changes remains elusive. In this sense, the genus Mazama is recognized by its high level of karyotype diversity among species with similar morphology.
