We demonstrate sensitive detection of alpha protons of fully protonated proteins by solid-state NMR spectroscopy with 100-111 kHz magic-angle spinning (MAS). The excellent resolution in the Cα-Hα plane is demonstrated for 5 proteins, including microcrystals, a sedimented complex, a capsid and amyloid fibrils. A set of 3D spectra based on a Cα-Hα detection block was developed and applied for the sequence-specific backbone and aliphatic side-chain resonance assignment using only 500 μg of sample. These developments accelerate structural studies of biomolecular assemblies available in submilligram quantities without the need of protein deuteration.

It is known that ultrashort echo time (UTE) magnetic resonance imaging (MRI) sequences can detect signals from water protons but not collagen protons in short T2 species such as cortical bone and tendons. The extent of coordination-induced bond weakening in aquo and hydroxo ligands bonded to a molybdenum(III) center complexed by a dianionic, pentadentate ligand system was probed by reacting the known complex (BPzPy)Mo(III)-NTf, , with degassed water or dry lithium hydroxide. The aquo adduct was not observed, but two LiNTf-stabilized hydroxo complexes were fully characterized. Computational and experimental work showed that the O-H bond in these complexes was significantly weakened (to ≈57 kcal mol), such that these compounds could be used to form the diamagnetic, neutral terminal molybdenum oxo complex (BPzPy)Mo(IV)O, , by hydrogen atom abstraction using the aryl oxyl reagent ArO• (Ar = 2,4,6-tri--butylphenyl).

The 'Viroporin' family comprises a number of mostly small-sized, integral membrane proteins encoded by animal and plant viruses. Despite their sequence and structural diversity, viroporins share a common functional trend: their capacity to assemble transmembrane channels during the replication cycle of the virus. Their selectivity spectrum ranges from low-pH-activated, unidirectional proton transporters, to size-limited permeating pores allowing passive diffusion of metabolites.
