Creating hydrogel systems to mimic the extracellular matrix is often limited by their static nature. The use of a two-photon [2+2] cycloaddition of maleimide groups to structure surfaces, to create hydrogels, and add 3D modifications with sub-micrometer precision is reported. The absence of photoinitiators and usage of near-infrared light is promising for future in vivo studies.

Hanbury-Brown and Twiss (HBT) effect is the foundation for stellar intensity interferometry. However, it is a phase insensitive two-photon interference effect. Here we extend the HBT interferometer by mixing intensity-matched reference fields with the input fields before intensity correlation measurement.

Microthrombus formation is associated with COVID-19 severity; however, the detailed mechanism remains unclear. In this study, we investigated mouse models with severe pneumonia caused by SARS-CoV-2 infection by using our in vivo two-photon imaging system. In the lungs of SARS-CoV-2-infected mice, increased expression of adhesion molecules in intravascular neutrophils prolonged adhesion time to the vessel wall, resulting in platelet aggregation and impaired lung perfusion.

Targeted cell ablation is a powerful strategy for investigating the function of individual neurons within neuronal networks. Multiphoton ablation technology by a tightly focused femtosecond laser, with its significant advantages of noninvasiveness, high efficiency, and single-cell resolution, has been widely used in the study of neuroscience. However, the firing activity of the ablated neuron and its impact on the surrounding neurons and entire neuronal ensembles are still unclear.

Immobilizing organic chromophores within the rigid framework of metal-organic frameworks (MOFs) augments fluorescence by effectively curtailing molecular motions. Yet, the substantial interspaces and free volumes inherent to MOFs can undermine photoluminescence efficiency, as they partially constrain intramolecular dynamics. In this study, we achieved optimization of both one- and two-photon excited fluorescence by incorporating linkers into an interpenetrated tetraphenylethene-based MOF (TPE-MOF).

HClO is considered a potential contributing factor and biomarker of inflammatory bowel disease (IBD). Accurate monitoring of lysosomal HClO is important for further developing specific diagnostic and therapeutic schedules for IBD. However, only rare types of fluorescent probes have been reported for detecting HClO in IBD so far.
