Flow birefringence property of desmin filaments.

We have investigated the flow birefringence property and assembly process of desmin, a muscle specific intermediate protein. Solution of non-polar desmin filaments showed birefringence when aligned in the sheared flow. The amount of birefringence of desmin filaments was considerably lower when compared with that of F-actin solution. Assembly of desmin from soluble state was followed by the birefringence measurements. At any desmin concentrations examined, the degree of flow birefringence increased rapidly just after the addition of the assembly buffer and reached a saturated level within 30 min. The time to reach half-maximal values of flow birefringence slightly but definitely depended on the initial soluble desmin concentrations. The plotting of the initial velocity of the assembly against the soluble desmin concentrations showed a slope of 1.4. This result suggested that the assembly process detected by flow birefringence measurements followed second-order kinetics, and the process corresponded to the second step of the three stage model for type III intermediate filament assembly proposed by Herrmann and his colleagues; the annealing of unit length filaments into filaments.

Blood flow alters the scattering behavior of penetration light, causing instability in the polarization state to emerge at the underlying tissue during polarization-sensitive optical coherence tomography (PSOCT). We propose an eigen decomposition method to meet this challenge, where the static and dynamic scattering signals are separated for PSOCT to provide the polarization measurements of the tissue of interest that is located beneath the blood flow. Using flow phantoms made by Intralipid solution and 3D-printed birefringent material, we show the flow-induced effects on the measurements of sample birefringent properties of optical axis, phase retardation, and degree of polarization uniformity.

In this study, the concept of rheo-optics was applied to explore the flow birefringence induced by the stress components along the camera's optical axis since it is often overlooked in the traditional theories of photoelastic flow measurement. A novel aspect of this research is that it involved conducting polarization measurements on simple shear flows, specifically from a perspective in which a shear-velocity gradient exists along the camera's optical axis. In response to an investigation on the paths of changes in the crystallization and radial differences during the forming process of nascent fibers, in this study, we conducted numerical simulation and analyzed the changes in crystallization mechanical parameters and tensile properties through a fluid dynamics two-phase model. The model was based on the melt-spinning method focusing on melt spinning, the environment of POLYFLOW, and the method of joint simulation, coupled with Nakamura crystallization kinetics, including the development of process collaborative parameters, stretch-induced crystallization, viscoelasticity, filament cooling, gravity term, inertia, and air resistance. Finally, for nylon 6 BHS and CN9987 resin spinning, the model successfully predicted the distribution changes in temperature, velocity, strain rate tensor, birefringence, and stress tensor along the axial and radial fibers and obtained the variation pattern of fibers' crystallinity along the entire spinning process under different stretching rates.
