Unlabelled: Although tryptophan (Trp) is the largest and most structurally complex amino acid, it is the least abundant in the proteome. Its distinct indole ring and high carbon content enable it to generate various biologically active metabolites such as serotonin, kynurenine (Kyn), and indole-3-pyruvate (I3P). Dysregulation of Trp metabolism has been implicated in diseases ranging from depression to cancer.

The present study investigated the covalent binding behavior of the flavonoids, catechin, eriodictyol, luteolin and quercetin with β-lactoglobulin (βlg). Since the four flavonoids possess the identical A- and B-ring structures, effects of the C-rings on the properties of flavonoids and the corresponding semiquinones are revealed. Experimental methods including DLS and CD spectra indicated that with quercetin at room temperature did not induce aggregation of βlg, whilst binding with the other three flavonoids resulted in aggregation of βlg.

Tryptophan (Trp) metabolism is linked to health and disease, with indoleamine 2,3-dioxygenase 1 (IDO) being a key enzyme in its breakdown outside the liver. This process produces metabolites that influence metabolic and inflammatory responses. A distinctive feature of the gut is its involvement in three major Trp catabolic pathways: the IDO-driven kynurenine pathway, bacteria-produced indoles, and serotonin.
