Unlabelled: A series of different amino acid-bearing thieno[2,3-d]pyrimidine moiety was synthesized via green chemistry aspects incorporating water as a solvent to give 2a-f, which were then acidified to attain the target compounds 3-9. Moreover, a tricyclic imidazothienopyrimidine of the glycine derivative (3) was synthesized to give (10). The title compounds were characterized by FT-IR, Mass, C-HNMR spectroscopy and microanalysis. All the obtained amino acid-derivatives were screened for their post-irradiation protective efficacy in young rats. γ-Irradiation exposure was employed to induce oxidative stress. Radiation triggered significant alterations in the hematologic and blood - biochemical parameters. Further, a significant deterioration of hepatic antioxidant status was observed. Furthermore, a significant elevation of Nuclear Factor-kappa B (NF-κB) protein expression and level, which induced elevation in Cyclooxygenase-2 (COX-2) activity and cytochrome P450 2E1 (CYP2E1) gene expression were detected in hepatic tissues. Additionally, elevated levels of Tumor Necrosis Factor-alpha (TNF-α), and Interleukin-6 (IL-6) were perceived in serum of γ-irradiated young rats. However, most of the newly synthesized derivatives showed significant protective effects against injuries induced by γ-irradiation exposure, via ameliorating the altered hematopoietic system and activity of different biochemical parameters in blood. The results indicate that the antioxidant and anti-inflammatory mechanism of these compounds could be attributed to the significant down-regulation NF-κB protein expression in hepatic tissues. Subsequently, NF-κB could regulate TNF-α and IL-6 levels, COX-2 activity and CYP2E1 gene expression. Methionine derivative 8 was found to be the most active one.
Conclusion: These new amino acid-derivatives showed promising outcomes as curative agents against γ-irradiation induced oxidative stress and physiological disturbance in different organs of young animals. Beyond, they may represent a novel selective class for treating liver injury induced by γ-irradiation in young rats, via down-regulation of NF-κB expression. Further investigations are warranted prior to the clinical use of these new compounds.

Skin flap transplantation is a conventional wound repair method in plastic and reconstructive surgery, but infection and ischemia are common complications. Photobiomodulation (PBM) therapy has shown promise for various medical problems, including wound repair processes, due to its capability to accelerate angiogenesis and relieve inflammation. This study investigated the effect of red and blue light on the survival of random skin flaps in methicillin-resistant Staphylococcus aureus (MRSA)-infected Sprague Dawley (SD) rats.

Experiments measuring evoked potentials require flexible and rapid adjustment of stimulation and recording parameters. In this study, we have developed a recording system and an associated Android application that allow making such adjustments wirelessly. The system consists of 3 units: for stimulation, recording and control.

Background And Purpose: Eukaryotic elongation factor 2 kinase (eEF2K) belongs to the Ca/calmodulin-dependent protein kinase family. We previously revealed that A484954, a selective eEF2K inhibitor, caused hypotensive and diuretic effects via the production of nitric oxide (NO) in spontaneously hypertensive rats. Otsuka Long-Evans Tokushima Fatty (OLETF) rats are hypertensive because of obesity and type 2 diabetes.

Orchidectomy and estrogenization of the male represent a procedure that is applicable in sex reassignment or in prostate cancer therapy. This approach has an influence on the hypothalamic-pituitary-adrenal axis and thus affects cardiovascular function and metabolism. We utilized orchidectomized rats to evaluate the effects of estradiol on the structure and hormonal output of the adrenal gland.

To examine the effect of DBS of the lateral hypothalamic area (LHA) on age-related memory changes, neuronal firing from CA1, oxidative stress, and the expression of Hsp70, BDNF, and synaptophysin. 72 male rats were randomly allocated into 6 equal groups: a) normal young group (8 W), b) sham young group, c) DBS young group, d) normal old group (24 months), e) sham old group and f) DBS old group. Memory tests (passive avoidance and Y maze), oxidative stress markers (MDA, catalase, and GSH) and expression of Nrf2, HO-1, Hsp70, BDNF, and synaptophysin were measured by the end of the experiment.
