Published: November 2016

Background: C. pseudotuberculosis is an important animal pathogen that causes substantial economical loss in sheep and goat farming. Zoonotic infections in humans are rare, but when they occur they are often severe and difficult to treat. One of the most studied proteins from this bacterium, the secreted protein CP40 is being developed as a promising vaccine candidate and has been characterized as a serine protease. In this study we have investigated if CP40 is an endoglycosidase rather than a protease.

Results: CP40 does not show any protease activity and contains an EndoS-like family 18 of glycoside hydrolase (chitinase) motif. It hydrolyzes biantennary glycans on both human and ovine IgGs. CP40 is not a general chitinase and cannot hydrolyze bisecting GlcNAc.

Conclusion: Taken together we present solid evidence for re-annotating CP40 as an EndoS-like endoglycosidase. Redefining the activity of this enzyme will facilitate subsequent studies that could give further insight into immune evasion mechanisms underlying corynebacterial infections in animals and humans.

Article Synopsis
  • Surgically treated pilonidal sinus disease (PSD) has high rates of wound complications, including surgical wound dehiscence, and this study aimed to see if negative pressure wound therapy (NPWT) can reduce these rates compared to conventional dressings.
  • A randomized controlled trial involved 50 patients aged 12-40 undergoing PSD excision, with equal splits between NPWT and conventional dressing; however, results showed no significant difference in wound dehiscence rates or recovery times between the two groups.
  • The study concluded that NPWT did not improve surgical outcomes for patients treated for PSD when compared to conventional passive dressings.

Department of Microbiology and Antimicrobial Resistance, Veterinary Research Institute, 62100 Brno, Czech Republic.

Caseous lymphadenitis (CLA) is a worldwide disease of small ruminants caused by , a facultative intracellular pathogen that is able to survive and multiply in certain white blood cells of the host. In this study, 33 strains of were isolated from sheep and goats suffering from CLA on nine farms in the Czech Republic. All these strains were tested for their antibiotic susceptibility, ability to form a biofilm and resistance to the effects of commonly used disinfectant agents.

Traditional Chinese medicine polysaccharides have numerous biological activities with broad applications in the biomedical industries. However, a clear understanding of the pharmacological activities of compound polysaccharides with multi-component structures remain challenging. This study aimed to investigate the immune boosting effect of compound polysaccharides on the influenza vaccine and assess the preliminary structure-activity relationship.

Background: Pulmonary arterial hypertension (PAH) is a severe cardiopulmonary disease characterized by complement dependent and proinflammatory activation of macrophages. However, effective treatment for complement activation in PAH is lacking. We aimed to explore the effect and mechanism of CP40-KK (a newly identified analog of selective complement C3 inhibitor CP40) in the PAH model.

Mapping B and T cell epitopes constitutes an important action for peptide vaccine design. PLD and CP40 virulence factors of biovar , a causal agent of Caseous Lymphadenitis, have been evaluated in a murine model as good candidates for vaccine development. Therefore, the goal of this work was to in silico analyze B and T cell epitopes of the PLD and CP40 proteins of a Mexican isolate of .
