Mitochondrial aspartate aminotransferase (GOT2) protein as a potential cryodamage biomarker in rooster spermatozoa cryopreservation.

Poult Sci

December 2024

State Key Laboratory of Animal Biotech Breeding, Institute of Animal Science, Chinese Academy of Agricultural Sciences, Beijing 100193, China. Electronic address:

Spermatozoa cryopreservation has been widely used for animal genetic conservation. Despite advances in chicken semen cryopreservation, the mechanism of spermatozoa cryodamage remains to be revealed. The cryopreservation process induces motion parameter decreased, structure damaged, proteomic and antioxidant system remodeled in spermatozoa.

Proteomic Analysis of Biological Fluids.

Methods Mol Biol

December 2024

Department of Environmental Toxicology, The University of California, Davis, Davis, CA, USA.

Biological fluids are proteinaceous liquids or suspensions released through different body orifices or through penetration of the skin. These fluids are the result of multiple tissues and cell types and contain extensive, highly complex, and dynamic protein populations that reflect both the transcriptional program of the originating cells and a record of the individual's health status. Body fluids are readily accessible to clinicians and researchers, and as such proteomic analyses are an important component of clinical studies, fertility studies, oral health studies, and forensic investigations.

Seminal fluid proteins in the Queensland fruit fly: Tissue origins, effects of mating and comparative genomics.

Insect Biochem Mol Biol

December 2024

Applied BioSciences, Macquarie University, NSW, 2109, Australia; CSIRO Environment, Black Mountain, ACT, 2601, Australia. Electronic address:

In many insect species, the ability of males to inhibit their mates from remating is an important component of fitness. This ability is also essential for the effective management of insect pests, including tephritid fruit flies, using the Sterile Insect Technique. Here we apply transcriptomics and proteomics to male reproductive tissues before and after mating to characterize components of semen that might mediate remating inhibition in Queensland fruit fly.

Top-down proteomics was employed to construct proteoform atlas of sperm and seminal plasma (SP) from bulls with low (LF) and high (HF) semen freezability. Sperm and seminal proteins were fractionated by tandem size exclusion chromatography (< 30 kDa) and analyzed by reversed-phase liquid chromatography-tandem mass spectrometry. This approach enabled the identification of 299 SP (from 46 families) and 267 sperm proteoforms (from 139 families).

Results of artificial insemination (AI) are affected by changes in sperm quality and the function throughout collection and preservation procedures. Proteome and metabolome alterations of sperm treated with the different procedures in goat, however, aren't fully understood. To this end, we sought to investigate the impacts of rectal probe electrostimulation (EE) and artificial vagina (AV) semen collection methods on the quality and the cryotolerance of goat sperm, with additional focus on proteomic and metabolomic analyses.
