Ynamides as Racemization-Free Coupling Reagents for Amide and Peptide Synthesis.

A highly efficient, two-step, one-pot synthetic strategy for amides and peptides was developed by employing ynamides as novel coupling reagents under extremely mild reaction conditions. The ynamides not only are effective for simple amide and dipeptide synthesis but can also be used for peptide segment condensation. Importantly, no racemization was detected during the activation of chiral carboxylic acids. Excellent amidation selectivity toward amino groups in the presence of -OH, -SH, -CONH, ArNH, and the NH of indole was observed, making the protection of these functional groups unnecessary in amide and peptide synthesis.

A straightforward synthetic route towards DAB-1 scaffolded dimeric iminosugars is described here, starting from readily available bis-glycosylamines. The method allows the integration of a variety of linkages (aryl, alkyl, polyethyleneglycol chains) between both iminosugars through the choice of the bis-amine used in the first step. Moreover, an additional substituent (allyl, ethynyl) may be inserted into the structure via nucleophilic addition of an organometallic reagent to the starting bis-glycosylamine.

: Over the past 40 years since the discovery of regenerating family proteins (Reg proteins), numerous studies have highlighted their biological functions in promoting cell proliferation and resisting cell apoptosis, particularly in the regeneration and repair of pancreatic islets and exocrine glands. Successively, short peptides derived from Reg3δ and Reg3α have been employed in clinical trials, showing favorable therapeutic effects in patients with type I and type II diabetes. However, continued reports have been limited, presumably attributed to the potential side effects.

Arsenic speciation in freshwater fish is crucial for providing meaningful consumption guidelines that allow the public to make informed decisions regarding its consumption. While marine fish have attracted much research interest due to their higher arsenic content, research on freshwater fish is limited due to the challenges in quantifying and identifying arsenic species present at trace levels. We describe here a sensitive method and its application to the quantification of arsenic species in freshwater fish.

We report here arsenic speciation in 1643 freshwater fish samples, representing 14 common fish species from 53 waterbodies in Alberta, Canada. Arsenic species were extracted from fish muscle tissue. Arsenic species in the extracts were separated using anion-exchange high-performance liquid chromatography (HPLC) and quantified using inductively coupled plasma mass spectrometry (ICPMS).
