SimC7 is a polyketide ketoreductase involved in biosynthesis of the angucyclinone moiety of the gyrase inhibitor simocyclinone D8 (SD8). SimC7, which belongs to the short-chain dehydrogenase/reductase (SDR) superfamily, catalyzes reduction of the C-7 carbonyl of the angucyclinone, and the resulting hydroxyl is essential for antibiotic activity. SimC7 shares little sequence similarity with characterized ketoreductases, suggesting it might have a distinct mechanism. To investigate this possibility, we determined the structures of SimC7 alone, with NADP(+), and with NADP(+) and the substrate 7-oxo-SD8. These structures show that SimC7 is distinct from previously characterized polyketide ketoreductases, lacking the conserved catalytic triad, including the active-site tyrosine that acts as central acid-base catalyst in canonical SDR proteins. Taken together with functional analyses of active-site mutants, our data suggest that SimC7 catalyzes a substrate-assisted, two-step reaction for reduction of the C-7 carbonyl group involving intramolecular transfer of a substrate-derived proton to generate a phenolate intermediate.

Preservation and shelf-life extension are of paramount importance for dried goji berry, especially those dehydrated using pulsed vacuum drying (PVD), which are generally applied for the high-quality dried products in the markets. This study compared the storage stability of dried goji berry dehydrated using PVD versus conventional hot wind drying (HWD), focusing on the complex interactions between packaging conditions and physicochemical compounds during accelerated storage. The results showed that PVD-dried goji berry exhibited higher browning degree (2.

Acoustic cavitation is a cutting-edge and eco-friendly advanced oxidation technology with significant efficacy in removing organic pollutants from water. Despite its potential, research on the degradation of o-cresol, a common and challenging phenolic pollutant, is limited. This study systematically investigates the optimal conditions for degrading o-cresol via acoustic cavitation and evaluates its application potential through extensive pilot tests.

In the presence of warm MeOH/DMSO (4:1), changed to {K(HO)}[(VO)Hdar(nah)]DMSO (·DMSO). Single crystal XRD studies of and confirm a binuclear structure along with a distorted square pyramidal geometry of each vanadium center where bis{ONO(2-)} ligands coordinate through phenolate-O, azomethine-N, and enolate-O atoms of each unit.

The reduction of carboxylic esters to aldehydes and alcohols is a fundamental functional group transformation in chemistry. However, the inertness of carbonyl group and the instability of ketyl radical anion intermediate impede the reduction of carboxylic esters via photochemical strategy. Herein, we described the reduction of aliphatic carboxylic esters with synergistic dual photocatalysis via phenolate-catalyzed single electron transfer process and thiol-catalyzed hydrogen atom transfer process.

Ferrate is a promising oxidizing agent for water treatment. Understanding the reaction characteristics and transformation mechanism of high-valent intermediate irons [Fe(V) and Fe(IV)] remains challenging. Here, we systematically investigated the roles of Fe(VI), Fe(V), and Fe(IV) species for acetaminophen oxidation using reaction kinetics, products, and stoichiometries.
