The NADPH oxidases are important transmembrane proteins producing reactive oxygen species (ROS). Within the Nox family, different modes of activation can be discriminated. Nox1-3 are dependent on different cytosolic subunits, Nox4 seems to be constitutively active and Nox5 is directly activated by calcium. With the exception of Nox5, all Nox family members are thought to depend on the small transmembrane protein p22phox. With the discovery of the CRISPR/Cas9-system, a tool to alter genomic DNA sequences has become available. So far, this method has not been widely used in the redox community. On such basis, we decided to study the requirement of p22phox in the Nox complex using CRISPR/Cas9-mediated knockout. Knockout of the gene of p22phox, CYBA, led to an ablation of activity of Nox4 and Nox1 but not of Nox5. Production of hydrogen peroxide or superoxide after knockout could be rescued with either human or rat p22phox, but not with the DUOX-maturation factors DUOXA1/A2. Furthermore, different mutations of p22phox were studied regarding the influence on Nox4-dependent HO production. P22phox Q130* and Y121H affected maturation and activity of Nox4. Hence, Nox5-dependent O production is independent of p22phox, but native p22phox is needed for maturation of Nox4 and production of HO.

Introduction: Chronic granulomatous disease is a defect in phagocytosis due to deficiency of gp91phox, p22phox, p47phox, p40phox, and p67phox (classic form of the disease). Recently, EROS and p40phox deficiency were described as responsible for the non-classical form of the disease. The 1,2,3-dihydrorhodamine oxidation technique, with phorbol-12-myristate-13-acetate as a stimulus, is performed to diagnose the classic chronic granulomatous disease.

LRRC8A drives NADPH oxidase-mediated mitochondrial dysfunction and inflammation in allergic rhinitis.

J Transl Med

November 2024

Department of Otolaryngology-Head and Neck Surgery, Shandong Provincial ENT Hospital, Shandong University, Jinan, 250000, Shandong, China.

Objectives: Allergic rhinitis (AR) is a complex disorder with variable pathogenesis. Increasing evidence suggests that the LRRC8A is involved in maintaining cellular stability, regulating immune cell activation and function, and playing significant roles in inflammation. However, the involvement of LRRC8A in AR inflammation and its underlying mechanisms remain unclear.

Article Synopsis
  • In Parkinson's disease (PD), microglia play a key role in maintaining cell health, and cycloastragenol (CAG) has potential neuroprotective effects by reducing inflammation.
  • This study aimed to investigate CAG's impact on microglial inflammasome activation and its mechanisms in PD treatment using mouse models and various laboratory techniques.
  • Results showed that CAG decreased neuroinflammation by promoting autophagy and reducing harmful reactive oxygen species, thus improving neuron health and behavior in PD mice, highlighting its therapeutic promise.

Article Synopsis
  • Mitochondrial dysfunction leads to increased production of reactive oxygen species (ROS), which is linked to various degenerative diseases like osteoarthritis.
  • Researchers developed a new treatment called "Mitocelle," a nanozyme made from biocompatible materials that inhibits the NOX4-p22phox interaction, which contributes to mitochondrial dysfunction.
  • In tests on a mouse model of osteoarthritis, Mitocelle demonstrated effective uptake into cartilage, long-term stability, and the ability to reduce joint damage, suggesting it could be a promising therapeutic option for related diseases.
