The pro-inflammatory leukotrienes (LTs) are formed from arachidonic acid (AA) in activated leukocytes, where 5-lipoxygenase (5-LO) translocates to the nuclear envelope to assemble a functional complex with the integral nuclear membrane protein 5-LO-activating protein (FLAP). FLAP, a MAPEG family member, facilitates AA transfer to 5-LO for efficient conversion, and LT biosynthesis critically depends on FLAP. Here we show that the novel LT biosynthesis inhibitor BRP-187 prevents the 5-LO/FLAP interaction at the nuclear envelope of human leukocytes without blocking 5-LO nuclear redistribution. BRP-187 inhibited 5-LO product formation in human monocytes and polymorphonuclear leukocytes stimulated by lipopolysaccharide plus N-formyl-methionyl-leucyl-phenylalanine (IC=7-10nM), and upon activation by ionophore A23187 (IC=10-60nM). Excess of exogenous AA markedly impaired the potency of BRP-187. Direct 5-LO inhibition in cell-free assays was evident only at >35-fold higher concentrations, which was reversible and not improved under reducing conditions. BRP-187 prevented A23187-induced 5-LO/FLAP complex assembly in leukocytes but failed to block 5-LO nuclear translocation, features that were shared with the FLAP inhibitor MK886. Whereas AA release, cyclooxygenases and related LOs were unaffected, BRP-187 also potently inhibited microsomal prostaglandin E synthase-1 (IC=0.2μM), another MAPEG member. In vivo, BRP-187 (10mg/kg) exhibited significant effectiveness in zymosan-induced murine peritonitis, suppressing LT levels in peritoneal exudates as well as vascular permeability and neutrophil infiltration. Together, BRP-187 potently inhibits LT biosynthesis in vitro and in vivo, which seemingly is caused by preventing the 5-LO/FLAP complex assembly and warrants further preclinical evaluation.

This work presents a novel application of additive manufacturing in the design of self-assembling helical viral capsids using 3D-printed components. Expanding on prior work with 3D-printed self-assembling spherical capsids, we developed helical models that integrate geometric parameters and magnetic interactions to mimic key features of the assembly process of helical viral capsids. Using dual-helix phyllotactic patterns and simplified electrostatic simulations, these models consistently self-assemble into a cylinder, providing unique insights into the structural organization and stability of helical capsids.

In this study, novel anion photo-responsive supramolecular hydrogels based on cysteine-silver sol (CSS) and iodate anions (IO) were prepared. The peculiarities of the self-assembly process of gel formation in the dark and under visible-light exposure were studied using a complex of modern physico-chemical methods of analysis, including viscosimetry, UV spectroscopy, dynamic light scattering, electrophoretic light scattering, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and X-ray photoelectron spectroscopy. In the dark phase, the formation of weak snot-like gels takes place in a quite narrow IO ion concentration range.

Taking into account the trends in the field of green chemistry and the desire to use natural materials in biomedical applications, (bio)polyelectrolyte complexes ((bio)PECs) based on a mixture of chitosan and gelatin seem to be relevant systems. Using the approach of self-assembly from the dispersion of the coacervate phase of a (bio)PEC at different ratios of ionized functional groups of chitosan and gelatin (), hydrogels with increased resistance to mechanical deformations and resorption in liquid media were obtained in this work in comparison to a hydrogel from gelatin. It was found that at ≥ 1 a four-fold increase in the elastic modulus of the hydrogel occurred in comparison to a hydrogel based on gelatin.

HIV-1 unspliced RNA serves two distinct functions during viral replication: it is packaged into particles as the viral genome, and it is translated to generate Gag/Gag-Pol polyproteins required for virus assembly. Recent studies have demonstrated that in cultured cells, HIV-1 uses multiple transcription start sites to generate several unspliced RNA species, including two major transcripts with three and one 5' guanosine, referred to as 3G and 1G RNA, respectively. Although nearly identical, 1G RNA is selected over 3G RNA to be packaged as the virion genome, indicating that these RNA species are functionally distinct.
