Histone deacetylase inhibitors (HDI) are promising anticancer therapies; however, drug resistance limits their efficacy. Here, we investigated the molecular mechanisms underlying HDI resistance, focusing on the mechanism of HDI-mediated induction of insulin-like growth factor 2 (IGF2) based on our previous study. The methylation status of CCCTC-binding factor (CTCF)-binding sites in the imprinting control region (ICR) were determined by methylation-specific PCR and bisulfite sequencing. The effectiveness of single or combinatorial blockade of DNA methyltransferase 1 (DNMT1) and histone deacetylase (HDAC) was evaluated using cell viability assay and patient-derived tumor xenograft (PDX) model. HDAC inhibition by vorinostat increased acetylated STAT3 (K685), resulting in transcriptional upregulation of DNMT1-mediated hypermethylation of CTCF-binding sites in the ICR decreased CTCF insulator activity, leading to a transcriptional upregulation of and activation of the insulin-like growth factor 1 receptor (IGF-1R) pathway in cells with acquired or vorinostat resistance. Strategies targeting DNMT1 diminished the expression and potentiated vorinostat sensitivity in preclinical models of lung cancer with hypermethylation in the ICR. The degree of ICR hypermethylation correlated with vorinostat resistance in patient-derived lung tumors and in patients with hematologic malignancies. DNMT1-mediated transcriptional upregulation of is a novel mechanism of resistance to HDIs, highlighting the role of epigenetic deregulation of IGF2 in HDI resistance and the potential value of the ICR hypermethylation and DNMT1 expression as predictive biomarkers in HDI-based anticancer therapies. .

This study evaluated the effect of wheat germ oil (WGO), Bacillus subtilis, and their combination on growth performance, immune response, nutrient digestibility, intestinal microbial, oxidative status, and gene expression in heat-stressed broilers. Four hundred one-day-old male Ross 308 broilers were distributed into five pens (20 birds/pen) in four experimental groups: a control (CON) without additives, WGO group fed diet with WGO at 200 mg.kg, BS group fed diet with B.

Autism spectrum disorder (ASD) is a pervasive neurobehavioral condition characterized by disruption of behavioral and emotional patterns in individuals with this condition. Given that various environmental and genetic factors play a fundamental role in the pathophysiology of ASD, it can be said that ASD is a multifaceted disease. This study used scientific databases including Google Scholar, PubMed, Scopus, and Semantic Scholar.

Background: Alzheimer's disease (AD) is a progressive neurodegenerative disease characterized by synapse and memory failure, and severe cognitive impairment. Physical exercise stimulates neuroprotective pathways, has pro-cognitive actions, and has been reported to alleviate memory impairment in AD. Irisin, an exercise-induced hormone, is secreted following proteolytic cleavage of fibronectin type-III-domain-containing 5 (FNDC5).

Background: Abnormal brain insulin signaling has been associated with Alzheimer's disease pathology and a faster rate of late-life cognitive decline. However, the underlying mechanisms remain unclear. In this study, we examined whether AD-related cortical proteins identified using targeted-proteomics play a role in the association of brain insulin signaling and cognitive decline.

Objectives: To determine the association between IGF2BP2 polymorphisms and type 2 diabetes mellitus.
Methods: This study involved 422 individuals, 214 diabetes mellitus cases, and 208 healthy controls. The PCR-RFLP technique was used to determine the genotype of the IGF2BP2 gene for the SNPs rs4402960 (G>T) and rs1470579 (A>C).
