Immunoelectron Microscopy of Cryofixed and Freeze-Substituted Plant Tissues.

Cryofixation and freeze-substitution techniques provide excellent preservation of plant ultrastructure. The advantage of cryofixation is not only in structural preservation, as seen in the smooth plasma membrane, but also in the speed in arresting cell activity. Immunoelectron microscopy reveals the subcellular localization of molecules within cells. Immunolabeling in combination with cryofixation and freeze-substitution techniques provides more detailed information on the immunoelectron-microscopic localization of molecules in the plant cell than can be obtained from chemically fixed tissues. Here, we introduce methods for immunoelectron microscopy of cryofixed and freeze-substituted plant tissues.

HtpB, the chaperonin of the bacterial pathogen , is found in extracellular locations, even the cytoplasm of host cells. Although chaperonins have an essential cytoplasmic function in protein folding, HtpB exits the cytoplasm to perform extracellular virulence-related functions that support 's lifestyle. The mechanism by which HtpB reaches extracellular locations is not currently understood.

Background: Chronic neuropathic pain generally has a poor response to treatment with conventional drugs. Extracellular vesicles released by the protozoan parasite display immunomodulatory properties towards mammalian immune cells. In this study, we have evaluated the potential of extracellular vesicles derived from the non-pathogenic protozoan towards the development of a vaccine adjuvant. As a proof of concept, we expressed in a codon-optimized SARS-CoV-2 Spike protein fused to the secreted acid phosphatase signal peptide in the N-terminal and to a 6×-His stretch in the C-terminal.
