In a Surface Plasmon Resonance (SPR) experiment two key parameters are classically recorded: the time and the angle of SPR reflectivity. This paper brings into focus a third key parameter: SPR reflectivity. The SPR reflectivity is proved to be related to surface roughness changes. Practical investigations on (i) gold anodizing and (ii) polypyrrole film growth in presence of oxalic acid is detailed under potentiostatic conditions. These experimental results reveal the potential of using the SPR technique to investigate real-time changes both on the gold surface, but also in the gold film itself. This extends the versatility of the technique in particular as sensitive in-situ diagnostic tool.

The dengue virus (DENV) NS5 protein plays a central role in dengue viral RNA synthesis which makes it an attractive target for antiviral drug development. This study introduces a novel approach to enhance surface plasmon resonance (SPR) sensing using Kramers-Kronig (K-K) relations for phase extraction from reflection spectra. By applying the K-K relations, a phase shift sensitivity of 3400°/RIU is achieved improving SPR detection limits to 9 × 10 RIU, which is four times more sensitive than conventional methods for determining SPR peak shifts. This advancement enables more precise detection of refractive index changes in aqueous solutions, making it valuable for biosensing and environmental monitoring with spectrometers.

Surface plasmon resonance (SPR) biosensors are extensively utilized for analyzing molecular interactions due to their high sensitivity and label-free detection capabilities. Recent innovations in surface-sensitive biosensors with dielectric surfaces address the inherent limitations associated with traditional gold surfaces, such as thermal effects and biocompatibility issues, which can impede broader applications. This review examines state-of-the-art biosensor configurations, including total internal reflection, optical waveguide, photonic crystal resonators, Bloch surface wave biosensors, and surface electrochemical biosensors, which can enhance analyte signals and augment the molecular detection efficiency at the sensor interface.
