In brief: Myositis ossificans-a benign process-is characterized by calcification of a hematoma that resulted from a severe contusion or strain or repeated trauma. Diagnosis is based on radiographic evidence of a calcified mass along with a history of recent trauma to the affected area. Management should be directed at minimizing or preventing extension of the original injury. Rest, ice, compression, and elevation (the classic treatment for any soft-tissue injury) is recommended as initial therapy. Gentle, painless range-of-motion exercises may be prescribed to help restore mobility. Padding can be used to protect the affected area from additional trauma.

Brain-derived neurotrophic factor (BDNF) and tropomyosin receptor kinase B (TrkB) are known to contribute to both protective and pronociceptive processes. However, their contribution to neuropathic pain after spinal cord injury (SCI) needs further investigation. In a recent study utilizing TrkB mice, it was shown that systemic pharmacogenetic inhibition of TrkB signaling with 1NM-PP1 (1NMP) immediately after SCI delayed the onset of pain hypersensitivity, implicating maladaptive TrkB signaling in pain after SCI.

Following spinal cord injury (SCI), there is a short-lived recovery phase that ultimately plateaus. Understanding changes within the spinal cord over time may facilitate targeted approaches to prevent and/or reverse this plateau and allow for continued recovery. Untargeted metabolomics revealed distinct metabolic profiles within the injured cord during recovery (7 days postinjury [DPI]) and plateau (21 DPI) periods in a mouse model of severe contusion SCI.
