The plant terpene synthase (TPS) family is responsible for the biosynthesis of a variety of terpenoid natural products possessing diverse biological functions. TPSs catalyze the ionization and, most commonly, rearrangement and cyclization of prenyl diphosphate substrates, forming linear and cyclic hydrocarbons. Moreover, a single TPS often produces several minor products in addition to a dominant product. We characterized the catalytic profiles of Hyoscyamus muticus premnaspirodiene synthase (HPS) and compared it with the profile of a closely related TPS, Nicotiana tabacum 5-epi-aristolochene synthase (TEAS). The profiles of two previously studied HPS and TEAS mutants, each containing nine interconverting mutations, dubbed HPS-M9 and TEAS-M9, were also characterized. All four TPSs were compared under varying temperature and pH conditions. In addition, we solved the X-ray crystal structures of TEAS and a TEAS quadruple mutant complexed with substrate and products to gain insight into the enzymatic features modulating product formation. These informative structures, along with product profiles, provide new insight into plant TPS catalytic promiscuity.

Caves are a unique ecosystem that harbor diverse microorganisms, and provide a challenging environment to the dwelling microbial communities, which may boost gene expression and can lead to the production of inimitable bioactive natural products. In this study, we obtained 59 actinobacteria from four different caves located in Bahadurkhel, District Karak, Pakistan. On the basis of taxonomic characteristics, 30 isolates were selected and screened for secondary metabolites production and bioactivity profiling.

The Flinders sensitive line (FSL) rat is an accepted rodent model for depression that presents with strong face, construct, and predictive validity, thereby making it suitable to investigate novel antidepressant mechanisms. Background: In this pilot study, the diagnostic agreement for sleep biomarker-based neurodegenerative disorder (NDD) risk probabilities was evaluated in patients with Alzheimer's disease dementia (AD), Lewy body disease (LBD), mild cognitive impairment (MCI), and controls (CG) with a Mini-Mental State Exam scores ≥28.
Methods: Sleep biomarkers recorded with the Sleep Profiler (Advanced Brain Monitoring, Inc.) were used as inputs to a 4-class machine learning algorithm trained to assign NDD risk probabilities to AD=27, LBD=19, isolated REM sleep behavior disorder=15, and CG=58.

Background: The present study aimed to establish a panel of plasma proteins that are associated with cognitive performance and amyloid-β burden and establish their utility for distinguishing early Alzheimer's disease (AD) from healthy controls.
Method: Plasma from fifty patients with early AD (all amyloid-positive) and forty-nine healthy controls (HCs) were assayed using quantitative mass spectrometry. The weighted correlation network analysis (WGCNA) was used to explore the association between plasma protein levels and cognitive performance and filter out the high-performance plasma proteomic biomarker candidates.
