Objectives: To evaluate urinalysis parameters useful for identifying mixed cultures in urine culture using an automated urinary particle analyzer to assess quality indicators (QIs) for urine sample contamination.

Methods: A retrospective observational cross-sectional study was conducted with 2,527 urine samples from patients of a quaternary hospital in Brazil. Urine samples were processed simultaneously in Sysmex UF-5000 flow cytometry analyzer (urinalysis) and MALDI-TOF (culture).

Microbial infections of the mammary gland often cause mastitis, and it can lead to substantial economic losses within the dairy industry due to its direct negative impact on milk production and composition and the associated treatment costs. Somatic cell count has emerged as a critical indicator in monitoring udder health, and recently, the large-scale availability of differential cell count analysis potentially offers new insights into underlying physiological processes. Therefore, the main objective of this study was to estimate the variation of DSCC and SCC of individual quarter-level milk samples of cows according to 1) their intramammary infectious status; 2) parity of the cow; 3) quarter location; and 4) DIM at the time of sampling.

Coumarin and coumarin-thiosemicarbazone hybrids were synthesized and characterized by various techniques such as FT-IR, H NMR, C NMR, MALDI-TOF-MS spectroscopy, and single crystal X-Ray diffractometer (XRD). The photochemical and photophysical properties of the compounds, such as solvatochromism, solubility, and chemical reactivity, were analyzed using UV-vis spectroscopy in different solvents. Due to the potential biological activities of the synthesized compounds, their binding affinity and mechanisms with calf thymus DNA (ct-DNA) and bovine hemoglobin (BHb) were determined using several useful spectrophotometric and theoretical approaches such as UV-vis absorption and fluorescence spectroscopy, molecular docking, and density functional theory (DFT).
