In this chapter, we describe how to shield a patch of a cell membrane against extracellularly applied chemoattractant stimuli. Classical patch clamp methodology is applied to allow for controlled shielding of a membrane patch by measuring the seal resistivity. In Dictyostelium cells, a seal resistivity of 50 MΩ proved to be tight enough to exclude molecules from diffusing into the shielded membrane region. This allowed for separating a shielded and a non-shielded region of a cell membrane to study the spatiotemporal dynamics of intracellular chemotactic signaling events at the interface between shielded and non-shielded areas. The spatiotemporal dynamics of signaling events in the membrane was read out by means of appropriate fluorescent markers using laser scanning confocal microscopy.

The elemental imaging of laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) provides spatial information on elements and therefore can further investigate the growth or evolution processes of an analyte. However, the accurate determination of spatial information is limited by the decoupling between the elemental distribution and mass spectrometry signals. This phenomenon, which is more distinct when high-diffusion ablation cells are used, arises from the overlap of ablation and the transport dispersion of aerosols.

Lamb waves are widely used for defect detection in structural health monitoring, and various methods are developed for Lamb wave data analysis. This paper presents an unsupervised Adversarial Transformer model for anomalous Lamb wave pattern detection by analyzing the spatiotemporal images generated by a hybrid PZT-scanning laser Doppler vibrometer (SLDV). The model includes the global attention and the local attention mechanisms, and both are trained adversarially.
