Virtually all recombinant proteins are now prepared using fusion domains also known as "tags". The use of tags helps to solve some serious problems: to simplify procedures of protein isolation, to increase expression and solubility of the desired protein, to simplify protein refolding and increase its efficiency, and to prevent proteolysis. In this review, advantages and disadvantages of such fusion tags are analyzed and data on both well-known and new tags are generalized. The authors own data are also presented.

Glucagon-like peptide-1 (GLP-1) analogues have emerged as effective treatments due to their ability to regulate blood glucose levels and gastric emptying through central nervous signals involving hypothalamic receptors, such as leptin. To address the short plasma half-life of native GLP-1, a C-16 fatty acid was conjugated to lysine in the GLP-1 analogue sequence to enhance its longevity.

Transpeptidases are specialized enzymes that have evolved for site-selective modification of peptides and proteins at their backbone termini. Approaches for adapting transpeptidases to catalyze side chain modifications are substantially more restricted, and typically rely on large recognition tags or require specific reaction conditions that are not easily compatible with broader applications. The synthesis of chiral amines is of critical importance but still challenging. Here, we present a self-sufficient and reusable dual-enzyme nanoreactor for chiral amine synthesis, featuring Z-mediated site-specific immobilization of amine dehydrogenase (AmDH) and glucose dehydrogenase (GDH) onto mesoporous silica nanoflowers (MSN). Molecular dynamics simulations revealed that the Z tag was bound to MSN via electrostatic interactions, thus maintaining the fusion enzyme's active pocket accessibility and improving its catalytic performance.
