Highly efficient catalytic stereoselective hydrogenative desymmetrization reactions mediated by rhodium complexes derived from enantiopure phosphine-phosphite (P-OP) ligands are described. The highest performing ligand, which contains a TADDOL-derived phosphite fragment [TADDOL = (2,2-dimethyl-1,3-dioxolane-4,5-diyl)bis(diphenylmethanol)], presented excellent catalytic properties for the desymmetrization of a set of achiral 1,4-dienes, providing access to the selective formation of a variety of enantioenriched secondary and tertiary alcohols (six examples, up to 92% ee).

Gold(I)-catalyzed highly enantioselective [4 + 2] cycloadditions of 1,6-enynes were achieved by utilizing chiral bifunctional P,N ligand. A wide range of 1,6-enynes were converted to enantioenriched 5-6-6-fused tricyclic compounds under mild reaction condition (up to 99% ee). This chiral gold(I) complex was also employed in the first desymmetric cycloadditions of 1,6-diynes bearing single ester group at the tether (up to 93% ee), where 5-- pathway predominates over 6-- pathway.

Asymmetric enamine alkylation represents a powerful tool for stereoselective C-C bond formation; in contrast, the development of enantioselective enamine acylation remains elusive. Here, we report that a chiral phosphoric acid can render an in-situ-formed enamine to undergo a stereoselective intramolecular α-carbon acylation, providing an alternative approach for the synthesis of useful pyrrolinones and indolinones bearing tetrasubstituted stereocenters. Utilizing an effective integration of the desymmetrization strategy and bifunctional organocatalysis, the first example of enantioselective enamine acylation is achieved by employing readily available aminomalonic esters and cyclic ketones.
