A diastereoselective synthesis of (S)-3-(3-(3,5-dimethyl-1H-pyrazol-1-yl)phenyl)-4-((R)-3-(2-(5,6,7,8-tetrahydro-1,8-naphthyridin-2-yl)ethyl)pyrrolidin-1-yl)butanoic acid (1), a potential therapeutic agent for the treatment of Idiopathic Pulmonary Fibrosis, which is currently undergoing Phase I clinical trials is reported. The key steps in the synthesis involved alkylation of 2-methylnaphthyridine with (R)-N-Boc-3-(iodomethyl)-pyrrolidine, and an asymmetric Rh-catalysed addition of an arylboronic acid to a 4-(N-pyrrolidinyl)crotonate ester. The overall yield of the seven linear step synthesis was 8% and the product was obtained in >99.5% ee proceeding with 80% de. The absolute configuration of 1 was established by an alternative asymmetric synthesis involving alkylation of an arylacetic acid using Evans oxazolidinone chemistry, acylation using the resulting 2-arylsuccinic acid, and reduction. The absolute configuration of the benzylic asymmetric centre was established as (S).

Chemical investigation of the ethyl acetate extract of the endophytic fungus Aspergillus terreus AArEF2 found in the fresh leaves of Artemisia arborescens L. led to isolation of five previously undescribed butenolides, asperterreunolides A-E (1-5), along with the known metabolite butyrolactone IV (6). The structure elucidation of these metabolites was established by detailed spectroscopic analyses (1D, 2D NMR and HR-ESI-MS analyses).

Ortho-quinone methides (o-QMs), generated by oxidative dehydration of clavatol, are highly reactive intermediates in biosynthesis that give rise to a variety of clavatol-containing pseudonatural products (PNPs) in fungi through intra- and intermolecular nonenzymatic cyclization/addition reaction, and some compounds have significant biological activities. Amino acids play important roles in human pathology and physiology and the qualitative and quantitative determination of chiral amino acids in humans and mammals also has important impacts on the life sciences. Therefore, the introduction of artificial probes to assess the concentrations and enantiomeric compositions [ee = ([D] - [L])/([D] + [L])] of amino acids in aqueous solution is necessary in understanding certain biological processes and diagnosing and treating diseases. Herein, a bifunctional achiral coumarin probe (Br-Coumarin) is reported to determine the absolute configuration, ee value, and concentration of 16 amino acids in THF/HO = 1/4 solution at micromolar concentrations.
