Prostaglandin E receptor subtype 3 (EP3) antagonism may treat a variety of symptoms from inflammation to cardiovascular and metabolic diseases. Previously, most EP3 antagonists were large acidic ligands that mimic the substrate, prostaglandin E2 (PGE2). This manuscript describes the optimization of a neutral small molecule amide series with improved lipophilic efficiency (LipE) also known as lipophilic ligand efficiency (LLE) ((a) Nat. Rev. Drug Disc.2007, 6, 881; (b) Annu. Rep. Med. Chem.2010, 45, 380).

Synthetic cannabinoids (SCs), which are among the most widely abused new psychoactive substances, are much more potent and have greater efficacy than natural cannabis. SCs can be disguised in various ways and are commonly sold in the form of electronic cigarette oil. SCs belong to a large family with structures consisting of a core with substituents, linker, ring with substituents, and tail.

Article Synopsis
  • A needle trap device (NTD) was created using a specific framework bonded to silica nanoparticles to analyze halogenated hydrocarbons (HHCs) in the air through gas chromatography-flame ionization detection (GC-FID).
  • The study utilized advanced characterization techniques to determine optimal experimental conditions, significantly reducing the need for extensive testing, material use, and costs.
  • Results showed that the NTD-GC-FID method is reliable and accurate for sampling HHCs in workplace air, aligning well with existing standard procedures for environmental monitoring.

An amide-based covalent organic framework (COF) was successfully synthesized using the reaction between 1,3,5-trimesoyl chloride and ethylenediamine. The structure and morphology of the COF were characterized using Fourier-transform infrared spectra, nuclear magnetic resonance spectroscopy, X-ray diffraction, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and Brunauer-Emmett-Teller surface area analysis. The COF was employed as a solid-phase extraction adsorbent for the sampling and preconcentration of chlorophenols from industrial wastewater samples prior to high-performance liquid chromatography with ultraviolet detection.

Seasonal and pandemic-associated influenza strains cause highly contagious viral respiratory infections that can lead to severe illness and excess mortality. Here, we report on the optimization of our small-molecule inhibitor F0045(S) targeting the influenza hemagglutinin (HA) stem with our Sulfur-Fluoride Exchange (SuFEx) click chemistry-based high-throughput medicinal chemistry (HTMC) strategy. A combination of SuFEx- and amide-based lead molecule diversification and structure-guided design led to identification and validation of ultrapotent influenza fusion inhibitors with subnanomolar EC cellular antiviral activity against several influenza A group 1 strains.

Article Synopsis
  • - The study focuses on improving lithium-metal batteries (LMBs) by using a highly concentrated electrolyte strategy to enhance amide usage, which helps control lithium reactivity and improves lithium deposition.
  • - To reduce costs in electrolyte preparation, researchers propose a new approach using dilute amide electrolytes (1 m Li-salt concentration) combined with perfluoropolyethers and specific additives to create a stable anion-rich lithium solvation structure.
  • - The resulting optimized electrolyte with N,N-Dimethyltrifluoroacetamide shows excellent compatibility and performance in both lab and practical applications, marking a significant advancement for stable lithium deposition and enhanced battery performance in LMBs.
