Escherichia coli is a bacterial species found ubiquitously in the intestinal flora of animals, although pathogenic variants cause major public health problems. Aptamers are short oligonucleotides that bind to targets with high affinity and specificity, and have great potential for use in diagnostics and therapy. We used cell-based Systematic Evolution of Ligands by EXponential enrichment (cell-SELEX) to isolate four single stranded DNA (ssDNA) aptamers that bind strongly to E. coli cells (ATCC generic strain 25922), with Kd values in the nanomolar range. Fluorescently labeled aptamers label the surface of E. coli cells, as viewed by fluorescent microscopy. Specificity tests with twelve different bacterial species showed that one of the aptamers-called P12-31-is highly specific for E. coli. Importantly, this aptamer binds to Meningitis/sepsis associated E. coli (MNEC) clinical isolates, and is the first aptamer described with potential for use in the diagnosis of MNEC-borne pathologies.

The emergence of numerous SARS-CoV-2 variants, characterized by mutations in the viral RNA genome and target proteins, has presented challenges for accurate COVID-19 diagnosis. To address this, we developed universal aptamer probes capable of binding to the spike proteins of SARS-CoV-2 variants, including highly mutated strains like Omicron. These aptamers were identified through protein-based SELEX using spike proteins from three key variants (D614G-substituted Wuhan-Hu-1, Delta, and Omicron) and virus-based SELEX, known as viro-SELEX.

We report a low-cost, portable biosensor composed of an aptamer-functionalized nanoporous anodic aluminum oxide (NAAO) membrane and a commercial microcontroller chip-based impedance reader suitable for electrochemical impedance spectroscopy (EIS)-based sensing. The biosensor consists of two chambers separated by an aptamer-functionalized NAAO membrane, and the impedance reader is utilized to monitor transmembrane impedance changes. The biosensor is utilized to detect amodiaquine molecules using an amodiaquine-binding aptamer (OR7)-functionalized membrane.

Sulfaquinoxaline (SQX) is widely utilized in aquaculture and animal husbandry due to its broad antimicrobial spectrum and low cost. However, it is difficult to degrade, and there are relevant residues in the aquatic environment, which could be harmful to both the ecological environment and human health. As a new recognition molecule, the aptamer can be recognized with SQX with high affinity and specificity, and the aptamer is no longer adsorbed to AuNPs after binding to SQX, which weakens the catalytic effect of AuNPs.
