Classic congenital adrenal hyperplasia (CAH) due to 21-hydroxylase deficiency results in severe cortisol and aldosterone deficiency, leading to persistent adrenal stimulation and excess production of ACTH and adrenal androgens. This study aimed to assess the causal relationship between lipidome and female reproductive diseases (FRDs) using an advanced series of Mendelian randomization (MR) methods. This study utilized genome-wide association study (GWAS) summary statistics encompassing 179 lipidomes and six prevalent FRDs, namely polycystic ovary syndrome (PCOS), endometriosis, uterine fibroid, female infertility, uterine endometrial cancer, and ovarian cancer. The two-sample MR (TSMR) approach was employed to investigate the causal relationships, with further validation using false discovery rate (FDR) and multivariable MR (MVMR) methods.

Infertility is increasingly linked to obesity, especially visceral fat accumulation. The Body Roundness Index (BRI) provides a precise quantification of body and visceral fat but its implications for infertility risks and diagnostic value are yet to be fully determined. This study analyzed data from 1,305 married women aged 20-45 years, drawn from the National Health and Nutrition Examination Survey (NHANES).
