Schistosomiasis is a major neglected parasitic disease that affects more than 265 million people worldwide and for which the control strategy consists of mass treatment with the only available drug, praziquantel. In this study, a series of new benzohydroxamates were prepared as potent inhibitors of Schistosoma mansoni histone deacetylase 8 (smHDAC8). Crystallographic analysis provided insights into the inhibition mode of smHDAC8 activity by these 3-amidobenzohydroxamates. The newly designed inhibitors were evaluated in screens for enzyme inhibitory activity against schistosome and human HDACs. Twenty-seven compounds were found to be active in the nanomolar range, and some of them showed selectivity toward smHDAC8 over the major human HDACs (1 and 6). The active benzohydroxamates were additionally screened for lethality against the schistosome larval stage using a fluorescence-based assay. Four of these showed significant dose-dependent killing of the schistosome larvae and markedly impaired egg laying of adult worm pairs maintained in culture.

We have previously identified that infection induces a unique form of myeloid training that protects male but not female mice from high fat diet induced disease. Here we demonstrate that ovarian derived hormones account for this sex specific difference. Ovariectomy of females prior to infection permits metabolic reprogramming of the myeloid lineage, with BMDM exhibiting carbon source flexibility for cellular respiration, and mice protected from systemic metabolic disease.

Spinal schistosomiasis, a rare manifestation of schistosomal infection, can closely mimic the presentation of spinal cord tumors and pose significant diagnostic challenges. We present the case of a 12-year-old boy from northern Ethiopia who experienced progressive back pain, tingling sensations in his lower extremities, and intermittent fever. Initially referred with a presumptive diagnosis of myxopapillary ependymoma for pediatric hematology-oncology evaluation, his marked eosinophilia and history of swimming in local rivers raised suspicion for spinal schistosomiasis.

Background: Parasitic infections are known to suppress the cell mediated immunity that protects against tuberculosis. The status of parasitic infections among bacteriologically confirmed tuberculosis patients and their household contacts in Cameroon is not well established. This study aimed at reporting the status of parasitic infections in TB patients and their household contacts with keen interest in associated risk factors to disease exposure.

Human schistosomiasis is a chronic neglected tropical disease caused by blood flukes of the genus Schistosoma, infecting 250 million people worldwide, mostly in sub-Saharan Africa. Recently, thousands of cases have been reported in immigrants to non-endemic countries, including Italy. Serological screening is recommended but so far, no accurate point-of-care (POC) and lab-free test is available.

Praziquantel is currently the only effective treatment for schistosomiasis, but several limitations underscore the need for new therapeutic agents. Recent promising in vitro results with Artemisia species and the success of A. annua and its active compound artemisinin in treating parasitic infections warrant the need for further studies.
