Ptychographic imaging has gained popularity for its high resolving power and sensitivity as well as for its ability to map simultaneously the sample's complex-valued refractive index and the illumination. Yet, despite significant progress that allows for reliable practical implementation, some of the technique's fundamentals remain poorly understood, and oftentimes successful data acquisition is either overly conservative or relies more on experimenters experience than on rational data acquisition strategies. Here, we propose a theoretical framework of ptychography, which is based on Gabor's notion of decomposition into elementary signals and the concept of frames. We demonstrate how this framework can straightforwardly be used to derive sampling requirements or to provide arguments on how to optimize the ptychographic scan. More generally, our theoretical framework can serve as a bridge between the experimental technique and the rich and well established mathematical disciplines of wavelets decomposition and spectrogram analysis.

The linear functional analysis, historically founded by Fourier and Legendre (Fourier's supervisor), has provided an original vision of the mathematical transformations between functional vector spaces. Fourier, and later Laplace and Wavelet transforms, respectively defined using the simple and damped pendulum, have been successfully applied in numerous applications in Physics and engineering problems. However the classical pendulum basis may not be the most appropriate in several problems, such as biological ones, where the modelling approach is not linked to the pendulum.

Photolyase and cryptochrome belong to a group of structurally similar flavoproteins but with two distinct functions of DNA repair as a photoenzyme and signal transduction as a photoreceptor, respectively, under blue-light illumination. Here, we studied a recently discovered bifunctional cryptochrome (CraCRY) with focus on its repair of UV-induced pyrimidine-pyrimidone (6-4) photoproduct (6-4PP). We used femtosecond spectroscopy and site-directed mutagenesis to map out the critical elementary steps by following the dynamics of initial reactants, various intermediates, and final products.

Calcium ions (Ca) are an archetypical and most versatile second messenger in virtually all cell types. Inspired by the discovery of Ca sparks in the 1990s, vibrant research over the last three decades has unveiled a constellation of Ca microdomains as elementary events of Ca signaling and, more importantly, a digital-analog dualism as the system design principle of Ca signaling. In this brief review, we present a sketchy summary on advances in the field of sparkology, and discuss how the digital subsystem can fulfill physiological roles otherwise impossible for any analog system.
