The production of recombinant proteins using the baculovirus expression vector system (BEVS) in large-scale agitated bioreactors is discussed in this chapter. Detailed methods of the key stages of a batch process, including host cell growth, virus stock amplification and quantification, bioreactor preparation and operation, the infection process, final harvesting, and primary separation steps for recovery of the product are presented. Furthermore, methods involved with advanced on-line monitoring and bioreactor control, which have a significant impact on the overall process success, are briefly discussed.

Introduction Monitored anesthesia care (MAC) is an anesthesia management method in which anesthesiologists use the minimum necessary intravenous anesthetics to achieve sedation and pain relief while maintaining spontaneous breathing. The challenge for the anesthesiologist is to find the correct balance between inhibiting the patient's stress response to invasive treatment, maintaining intraoperative hemodynamic stability, and doing so as quickly as possible. We hypothesized that diluting remifentanil (D-remi) to 10 μg/mL and increasing the rate of administration would increase the responsiveness of the syringe pump and allow for better control of anesthetic depth.

The production and purification of the secreted ectodomain of SARS-CoV-2 spike protein (S protein) were performed by transiently transfecting suspension-adapted Chinese hamster ovary cells (ExpiCHO). The method involved the separate addition of plasmid DNA expressing the S protein and polyethyleneimine to a suspension culture at a density of 5 × 10 cells/mL; and the subsequent addition of dimethyl sulfoxide at 2% (v/v). The transfected ExpiCHO cells were cultivated at 31 °C with agitation by orbital shaking under 5% CO.

Cadmium (Cd), a carcinogen, is released from industrial activities like metal refineries and battery runoff, with significant contamination reported near zinc smelters in Korea. This study addresses the issue using an efficient, economical adsorption process with waste-derived biochar-based adsorbents known for high Cd removal. Poultry manure (PM), typically used as fertilizer, can lead to environmental pollution if mismanaged; therefore, it was pyrolyzed to produce biochar.
