Objective: Paraoxonase 1 (PON-1) is a high-density lipoprotein (HDL)-associated antioxidant enzyme that plays an important role in HDL-mediated cardioprotection. Although genetic polymorphisms are known to modulate PON-1 activity, its involvement in cardiovascular disease (CVD) in rheumatoid arthritis (RA) is controversial, suggesting that other factors may modulate its function. Since anti-HDL antibodies have been found to be related to an impaired lipid profile and occurrence of CVD in RA, this study was undertaken to examine the associations between PON-1 activity, anti-HDL antibodies, and CVD according to PON1 genetic variants in patients with RA.

Methods: Serum PON-1 activity, using paraoxon as substrate, and IgG anti-HDL antibodies were quantified in 212 RA patients and 110 healthy controls. The PON1 rs662 genotype (Q>R) was determined with TaqMan probes. An additional group of 13 biologics-naive patients with RA was prospectively followed up for 3 months.

Results: PON-1 activity was decreased in RA patients compared to healthy controls (P = 0.005), and an effect of the rs662 genotype was noted in both groups, with Q/Q homozygotes exhibiting the lowest PON-1 activity. The distribution of rs662 genotypes did not differ between RA patients and healthy controls (P = 0.215). In patients carrying the Q/Q genotype, anti-HDL antibodies were associated with impaired PON-1 activity (P = 0.010), and levels of anti-HDL antibodies were associated with decreased HDL levels (r = -0.680, P < 0.001) and higher prevalence of cardiovascular events, as determined in univariate and multivariate models. Furthermore, change in anti-HDL antibody levels upon tumor necrosis factor blockade was an independent predictor of improved PON-1 activity (β = -0.369, 95% confidence interval -0.669, -0.069; P = 0.024).

Conclusion: PON-1 activity is impaired in RA in association with the rs662 genotype and anti-HDL antibodies, the latter being recognized as a pivotal player in the link between rs662 and CVD in patients with RA.

Objective: Dysregulation of lipid metabolism can be one of the pathophysiological mechanisms linking high-density lipoprotein cholesterol (HDL-C) dysfunction to obesity. The aim of the study is to show possible changes in lipid metabolism with atherogenic indices in obese patients after sleeve gastrectomy (SG) surgery.

Method: Thirty patients who had SG surgery for obesity were included in the prospective study.

Objective: Therapeutic drug monitoring (TDM) indicators have been suggested to predict overall outcome responses to olanzapine (OLZ) treatments in terms of efficacy and metabolic syndrome. This study aimed to investigate whether paraoxonase-1 (PON-1) activity can be used to predict schizophrenia patient outcomes.

Methods: Schizophrenic patients ( = 50) aged between 20 and 65 years who received OLZ treatment were recruited, and their Positive and Negative Syndrome Scale scores, PON-1 activity, and olanzapine drug levels normalized by dose (OLZ/D) and its metabolite N-desmethyl-olanzapine (DMO), together with biochemical parameters, were determined.

Mast cell tumors (MCTs) are common skin neoplasms in dogs. Prognostic indicators include histologic grade, clinical stage, high Ki-67 index, elevated argyrophilic nucleolus organizer regions (AgNOR) index, mutations, and recurrence after surgery. Blood serum redox status has been shown to correlate with prognostic factors in canine lymphoma and mammary tumors.

Introduction: Paraoxonase-1 (PON1) is a crucial esterase in cardiovascular health, closely associated with HDL and known for its antioxidant and anti-inflammatory properties. Reduced PON1 activity has been linked to cardiovascular diseases. Lysophospholipids (LysoPLs), essential for cellular processes and immune responses, are implicated in the pathogenesis of cardiovascular diseases and are bound to lipoproteins, contributing to their diverse effects.

Article Synopsis
  • PCOS is a common syndrome in women that relates to issues like insulin resistance, and the study focuses on the effects of chlorogenic acid from green coffee on oxidative stress markers.
  • In a randomized clinical trial with 44 women, those taking 400 mg of green coffee for 6 weeks showed increased PON-1 levels and decreased cholesterol levels, indicating potential health benefits.
  • Despite the positive changes in PON-1 and cholesterol, the green coffee supplementation did not significantly affect levels of MDA or glycemic status in the participants.
