Background: FTIH studies can be challenging due to the varying dosing regimens and rapid data delivery. Chemists are asked to provide ultra-low limits of quantitation to provide an understanding of patient efficacy and safety in order to progress drug development. In a recent dermal study it became necessary to reduce the LLOQ of a small molecule drug from 50 to 1 pg/ml due to reductions in the dose and surface area of drug application.
Methodology: The 50-fold increase in assay sensitivity necessitated the use of a high-resolution mass spectrometer (LC-HRMS) to separate matrix interferences observed when using a unit resolution triple quadrupole MS.
Conclusion: A sensitive, robust assay was validated to support of a FTIH study using a LC-HRMS.

Short-chain enoyl-CoA hydratase 1 deficiency (ECHS1D) is a rare genetic disorder caused by biallelic pathogenic variants in the ECHS1 gene. ECHS1D is characterised by severe neurological and physical impairment that often leads to childhood mortality. Therapies such as protein and single nutrient-restricted diets show poor efficacy, whereas the development of new treatments is hindered by the low prevalence of the disorder and a lack of model systems for treatment testing.

Wolbachia, as a widely infected intracellular symbiotic bacterium in Arthropoda, is able to manipulate the reproduction of insect hosts for facilitating their own transmission. Cytoplasmic incompatibility (CI) is the most common phenotype that Wolbachia induced in insect hosts where they resulted in the failure of uninfected egg hatch when fertilized with the sperm derived from Wolbachia-infected males, suggesting that the sperm are modified by Wolbachia during spermatogenesis. Although the molecular mechanisms of CI are beginning to be understood, the effects of Wolbachia on the symbiotic relationship and the proper dynamics of spermatogenesis have not yet been fully investigated.

Larvae of the black soldier fly Hermetia illucens have potential as a natural waste recycler and subsequent use as protein-rich feed for livestock. A common question about the insect-farming processes is, what about the concerns of mass escape of insects from large populations? Here, we present a binary transgenic CRISPR/Cas9 system to generate wingless strain with the potential to address this issue. We identified gonad-specific promoters in vivo and evaluated use of the two strongest promoters, nanos and exuperantia, to drive Cas9 expression.

In recent years, global climate change has caused worldwide trends in science and industry toward a focus on the development of modern technologies with reduced environmental impact, including reduced CO emissions into the atmosphere. The technology for producing asphalt mixtures (AM) at lower temperatures (WMA-warm asphalt mix) using zeolite materials for the bitumen foaming process fits perfectly into these trends. Therefore, towards the development of this technology, the research presented in this paper presents the modification process of zeolite NaP1 from fly ash with silanes of different chemical structures (TEOS, MPTS, TESPT) and their application in the foaming process of bitumen modified with polymers (PMB 45/80-55).
