A doxorubicin (DOX)-loaded and light-induced ROS-producing polymeric micelle (D-LRPM), in which light triggers simultaneous DOX-release and endo/lysosomal escape, produces a powerful, spatiotemporally controllable, therapeutic efficacy for tumor treatment.

Monotherapy has poor accuracy and is easily restricted by tumor microenvironment (TME). Remodeling components of the TME to activate multimodal cancer therapy with high precision and efficiency is worth exploring. Ferroptosis, triggered by membrane lipid peroxidation (LPO) and diminished antioxidants, can be induced by intracellular iron (II, Fe). However, the role of nitric oxide (NO) in causing Fe overload for ferroptosis remains uncertain. Considering that the precise delivery of Celastrol (Cst) into mitochondria to induce mitochondrial dysfunction may be a potential approach to improve the therapeutic outcomes of Cst on TNBC, a novel tumor mitochondria dual-targeted mixed-micelle nano-system was fabricated via self-synthesized triphenylphosphonium-modified cholesterol (TPP-Chol) and hyaluronic acid (HA)-modified cholesterol (HA-Chol). The Cst-loaded mixed micelles (Cst@HA/TPP-M) exhibited the characteristics of a small particle size, negative surface potential, high drug loading of up to 22.8%, and sustained drug release behavior.

In the past decade, nucleic acid therapies have seen a boon in development and clinical translation largely due to advances in nanotechnology that have enabled their safe and targeted delivery. Nanoparticles can protect nucleic acids from degradation by serum enzymes and can facilitate entry into cells. Still, achieving endosomal escape to allow nucleic acids to enter the cytoplasm has remained a significant barrier, where less than 5% of nanoparticles within the endo-lysosomal pathway are able to transfer their cargo to the cytosol.
