Insulin-like peptides (ILPs) are functional analogs of insulin and have been identified in many insect species. The insulin cell signaling pathway is a conserved regulator of metabolism, and in insects, as well as in other animals, can modulate physiological functions associated with the metabolism of lipids and carbohydrates. In the present study, we have identified the first ILP from the Rhodnius prolixus genome (termed Rhopr-ILP) and investigated its involvement in energy metabolism of unfed and recently fed fifth instars. We have cloned the cDNA sequence and analyzed the expression profile of the transcript, which is predominantly present in neurosecretory cells in the brain, similar to other insect ILPs. Using RNAi, we have reduced the expression of this peptide transcript by 90% and subsequently measured the carbohydrate and lipid levels in the hemolymph, fat body and leg muscles. Reduced levels of Rhopr-ILP transcript induced increased carbohydrate and lipid levels in the hemolymph and increased lipid content in the fat body, in unfed insects and recently fed insects. Also their fat bodies displayed enlarged lipid droplets within the cells. On the other hand, the carbohydrate content in the fat body and in the leg muscles of unfed insects were decreased when compared to control insects. Our results indicate that Rhopr-ILP is a modulator of lipid and carbohydrate metabolism, probably through signaling the presence of available energy and nutrients in the hemolymph.

Behavior-change lifestyle interventions are fundamental in children and adolescent obesity management. This scoping review discusses optimal behavior-change lifestyle interventions in the treatment of overweight and obesity in children and adolescents. A literature search on diet, physical activity, and behavioral intervention for obesity treatment in children and adolescents aged 0-19 years was conducted in the Cochrane Library, MEDLINE (OVID), EMBASE, and ClinicalTrials.

Ethambutol is used to treat tuberculosis (TB) in individuals living with HIV. Low concentrations of ethambutol have been reported in patients dosed with the World Health Organization (WHO)-recommended first-line regimen. We analyzed the pharmacokinetics of ethambutol in 61 HIV-positive individuals diagnosed with drug-sensitive TB enrolled in the tuberculosis and highly active antiretroviral therapy (TB-HAART) study.

Recent sets of evidence have described profiles of 16S rDNA sequences in host tissues, notably in fat pads that are significantly overrepresented and can serve as signatures of metabolic disease. However, these recent and original observations need to be further detailed and functionally defined. Here, using state-of-the-art targeted DNA sequencing and discriminant predictive approaches, we describe, from the longitudinal FLORINASH cohort of patients who underwent bariatric surgery, visceral, and subcutaneous fat pad-specific bacterial 16SrRNA signatures.

Background: Postoperative complications are prone to occur in patients after radical pancreaticoduodenectomy (PD). This study aimed to construct and validate a model for predicting postoperative major complications in patients after PD.
Methods: The clinical data of 360 patients who underwent PD were retrospectively collected from two centers between January 2019 and December 2023.
