Orf virus (ORFV) is a zoonotic parapoxvirus that causes pustular dermatitis of sheep, and occasionally humans. Despite causing sustained infections, ORFV induces only a transient increase in pro-inflammatory signalling and the trafficking of innate immune cells within the skin seems to be impaired. An explanation for this tempered response to ORFV infection may lie in its expression of a homolog of the anti-inflammatory cytokine, interleukin (IL)-10. Using a murine model in which inflammation was induced by bacterial lipopolysaccharide, we examined the effects of the ORFV-IL-10 protein on immune cell trafficking to and from the skin. ORFV-IL-10 limited the recruitment of blood-derived Gr-1(int)/CD11b(int) monocytes, CD11c(+ve)/MHC-II(+ve) dendritic cells and c-kit(+ve)/FcεR1(+ve) mature mast cells into inflamed skin. ORFV-IL-10 also suppressed the activation of CD11c(+ve)/MHC-II(+ve) dendritic cells within the skin, reducing their trafficking to the draining lymph node. These findings suggest that expression of IL-10 by ORFV may contribute to the impaired trafficking of innate immune cells within infected skin.

Objectives: To investigate the pathogenicity of feline herpesvirus-1 (FHV-1) to the cornea, FHV-1 strains isolated from feline eyes with dendritic ulcers were subjected to genomic analysis to determine whether FHV-1 vaccine strains are involved in the formation of dendritic ulcers.
Methods: All open reading frame (ORF) sequences of the three F2 strains (Virbac, Intervet and Merial) and the FHV-1 clinical isolates from cats registered in GenBank were compared to detect nucleotide variants unique to the F2 strains, with those nucleotides then being used for simple genotyping of the F2 strains. In all isolates from feline eyes with dendritic ulcers, the regions including nucleotide variants of the F2 strain were amplified with PCR and sequenced.

The NS1 protein of nine mosquito-borne flaviviruses, including Dengue virus 1-4, Japanese encephalitis virus, West Nile virus, Yellow fever virus, Tembusu virus, and Zika virus, shows distinct codon usage and evolutionary traits. Codon usage analysis shows notable base composition bias and non-conservatism in NS1, with distinct evolutionary traits from its ORF. Analysis of relative synonymous codon usage (RSCU) indicates that the NS1 genes exhibit non-conservative RSCU patterns within different mosquito-borne pathogenic flaviviruses.
