The antitumor activity shown by many platinum complexes has produced a strong interest in research of new organometallic compounds having anticancer action. Among the many metal compounds synthesized and tested, those based on titanium have received considerable attention because of their cytotoxic activity against solid tumors. Particularly, new titanocene compounds containing aromatic groups linked to the Cp (cyclopentadienyl ring, C5H5) have been synthetized, such as the titanocene Y (bis-[(p-methoxybenzyl)cyclopentadienyl]titanium dichloride) that displayed promising medium-high cytotoxic activity on breast cancer cell lines. Other titanocene complexes recently synthesized, obtained by replacing the substituent methoxy-aryl of cyclopentadienes of titanocene Y with ethenyl-methoxide or ethenyl-phenoxide, showed increased cytotoxic activity on breast cancer cell lines being more stable compounds. In this paper, we report that new titanocene complexes holding lipophilic groups, for instance a methyl group on benzyl carbon, exhibit improved antiproliferative effect on breast cancer cell line MCF-7. Similar results have been obtained introducing a 5-methoxy naphthyl group to further stabilize the titanocene complexes. These inhibitory effects on breast cancer cells have been ascribed to human topoisomerase I and II inhibition as demonstrated by specific enzymatic assays.

Well-defined amorphous/semi-crystalline statistical copolymers of n-dodecyl isocyanate, DDIC, and allyl isocyanate, ALIC, were synthesized via coordination polymerization using the chiral half-titanocene complex CpTiCl(O-(S)-2-Bu) as an initiator. In the frame of the terminal model, the monomer reactivity ratios of the statistical copolymers were calculated using both well-known linear graphical methods and the computer program COPOINT. The molecular and structural characteristics of the copolymers were also calculated.

The reduction of titanocene dichloride CpTiCl with lanthanide metals has led to the discovery of a surprising lanthanide effect: while with most lanthanides, a divalent [CpTi] equivalent was obtained, the use of samarium or ytterbium only led to the reduction to trivalent [CpTiCl]-type complexes, including the structurally characterized heterobimetallic complex [CpTi(μ-Cl)SmCl(THF)]. These results were corroborated by reactivity studies (alkyne coupling and radical reactions), EPR spectroscopy and electrospray mass spectrometry, providing new insights into the reduction chemistry of lanthanide metals.

The preparation of polyolefins with high polar monomer contents (above 20 mol %) has long been a challenge. Half-titanocenes (Cp')[HCN(Ar)]BOTiCl bearing bulky electron-donating N-heterocyclic boryloxy ligands have been designed and synthesized. The complexes (Cp*)[HCN(Ar)]BOTiCl (2, Ar=2,6-iPrCH; 5, Ar=2,4,6-MeCH) supported by Cp* and the boryloxy ligands have been shown to efficiently catalyze the copolymerization of ethylene and long chain α-olefins.

Titanium, as an important transition metal, has garnered extensive attention in both industry and academia due to its excellent mechanical properties, corrosion resistance, and unique reactivity in organic synthesis. In the field of organic photocatalysis, titanium-based compounds such as titanium dioxide (TiO), titanocenes (CpTiCl, CpTiCl), titanium tetrachloride (TiCl), tetrakis(isopropoxy)titanium (Ti(OPr)), and chiral titanium complexes have demonstrated distinct reactivity and selectivity. This review focuses on the roles of these titanium compounds in photocatalytic organic reactions, and highlights the reaction pathways such as photo-induced single-electron transfer (SET) and ligand-to-metal charge transfer (LMCT).

After the discovery of cis-platin, the first metal-based anticancer drugs, budotitane, and titanocene dichloride entered clinical trials. These two classes of complexes were effective against those cell lines that are resistant to cis-platin and other platinum-based drugs.
