A ratiometric two-photon fluorescent probe was developed based on quinoline platform to detect hydrazine (NH) with high selectivity. exhibited large two-photon absorption cross sections at 710 nm (250 GM) and excellent ratiometric two-photon fluorescent detection signal for hydrazine. was also successfully applied to selectively detect hydrazine vapor even at a concentration down to 0.05%. Cell cytotoxicity and bio-imaging studies revealed that probe was cell-permeable and could be used to detect hydrazine in living cells with low cytotoxicity under two-photon excitation.

Alzheimer's disease (AD) is characterized by multiple toxicity from various biomarkers, and it is very important to monitor the fluctuation of biomarker level in brain tissues to track its early onset, illness progression, and therapeutic effect. Hypochlorous acid (HOCl) is confirmed to be a reliable biomarker to extend the scope of diagnosis. However, the practical applications of the developed conjugated small molecule fluorescent probes for detecting HOCl are often restricted to a large extent by their low solubility, aggregation-caused quenching (ACQ), unsatisfactory fluorescence brightness, small Strokes-shift, and lack of self-correcting ratiometric emitters.

Article Synopsis
  • The visualization of microRNA (miRNA) in cancer cells is crucial for understanding disease progression, and two-photon (TP) imaging offers distinct advantages over traditional methods due to its high resolution and deep tissue penetration.
  • This study introduces a two-photon silica nanoamplifier (TP-SNA) that combines TP dye-doped silica nanoparticles with a signal amplification strategy, enabling highly sensitive detection of endogenous miRNAs in tumor cells at various depths.
  • The TP-SNA successfully detected miR-203 with a detection limit of 33 pM and achieved a tissue penetration depth of 210 μm, showing potential for clinical diagnosis applications.

A luminescent nanoprobe capable of orthogonal sensing of two independent events is highly significant for unbiased disease-related detection such as the detection of senescent cells. Moreover, it is invaluable that the nanoprobe possesses a two-photon excitable characteristic that is highly suitable for imaging living cells and tissues. Herein, we present a two-photon-excitable multiluminescent orthogonal-sensing nanoprobe (OS nanoprobe) capable of detecting both pH elevation and β-galactosidase (β-gal) overexpression in senescent cells.

The dysregulation of pH has been linked to the onset of chronic conditions, such as cancer and neurological diseases. Consequently, the development of a highly sensitive tool for intracellular pH sensing is imperative to investigate the interplay between pH and the biochemical changes accompanying disease pathogenesis. Here, we present the development of a ratiometric fluorescent nanoprobe, , designed for precisely measuring pH levels.
