Apolipophorin III, a 163 residue monomeric protein from the greater wax moth Galleria mellonella (abbreviated as apoLp-IIIGM), has roles in upregulating expression of antimicrobial proteins as well as binding and deforming bacterial membranes. Due to its similarity to vertebrate apolipoproteins there is interest in performing atomic resolution analysis of apoLp-IIIGM as part of an effort to better understand its mechanism of action in innate immunity. In the first step towards structural characterization of apoLp-IIIGM, 99 % of backbone and 88 % of side chain (1)H, (13)C and (15)N chemical shifts were assigned. TALOS+ analysis of the backbone resonances has predicted that the protein is composed of five long helices, which is consistent with the reported structures of apolipophorins from other insect species. The next stage in the characterization of apoLp-III from G. mellonella will be to utilize these resonance assignments in solving the solution structure of this protein.

Peptides, due to their diverse and controllable properties, are used as both liquid and gas phase recognition elements for both biological and chemical targets. While it is well understood how binding of a peptide to a biomolecule can be converted into a sensing event, there is not the same mechanistic level of understanding with regard to how peptides modulate the selectivity of semiconductor/conductor-based gas sensors. Notably, a rational, mechanistic study has not yet been performed to correlate peptide properties to the sensor response for volatile organic compounds (VOCs) as a function of chemical properties.

Sum frequency generation vibrational spectroscopy was applied to study the surface hydration and protein adsorption behavior on several polymer coatings based on pyridine, imidazole, and amine side groups along with vinyl or methacrylate backbones and their corresponding zwitterionic forms with carboxybetaine or sulfobetaine side chains, prepared by initiated chemical vapor deposition (iCVD). iCVD also enables facile tuning of the cross-linking density of the polymer coatings by blending in a cross-linker during the deposition, namely, 1,3,5,7-tetramethyl-1,3,5,7-tetravinyl cyclotetrasiloxane. Our results show that both the low- and high-cross-linking density zwitterionic polymers exhibit significantly better antifouling activities compared to those of the polymers without the zwitterionic side chains.
