The Critical Assessment of protein Structure Prediction (CASP) experiment would not have been possible without the prediction targets provided by the experimental structural biology community. In this article, selected crystallographers providing targets for the CASP11 experiment discuss the functional and biological significance of the target proteins, highlight their most interesting structural features, and assess whether these features were correctly reproduced in the predictions submitted to CASP11. Background: Protein secondary structure prediction (PSSP) is a critical task in computational biology, pivotal for understanding protein function and advancing medical diagnostics. Recently, approaches that integrate multiple amino acid sequence features have gained significant attention in PSSP research.

Objectives: We aim to automatically extract additional features represented by evolutionary information from a large number of sequences while simultaneously incorporating positional information for more comprehensive sequence features.

Caspase-11 signaling promotes damage to hippocampal CA3 to enhance cognitive dysfunction in infection.

Mol Med

August 2024

Department of Physiology, School of Basic Medical Science, Central South University, Changsha, 410000, People's Republic of China.

Background: Cognitive dysfunction caused by infection frequently emerges as a complication in sepsis survivor patients. However, a comprehensive understanding of its pathogenesis remains elusive.

Methods: In our in vivo experiments, an animal model of endotoxemia was employed, utilizing the Novel Object Recognition Test and Morris Water Maze Test to assess cognitive function.

Article Synopsis
  • PKDL is a significant skin condition that develops after visceral leishmaniasis (VL), leading to social stigma due to skin lesions caused by the Leishmania donovani parasite.
  • Inflammation in both VL and PKDL can exacerbate tissue damage, allowing the parasite to persist while causing changes in skin color.
  • Recent research has analyzed inflammasomal markers and miRNA networks, suggesting that targeting these pathways could help develop treatments to prevent disease progression and its related social impacts.

Investigating the role of Caspase-1 in a mouse model of Juvenile X-linked Retinoschisis.

Front Med (Lausanne)

June 2024

Department of Ophthalmology and Visual Sciences, IVR, University of Iowa, Iowa City, IA, United States.

Purpose: Previous studies have reported Caspase-1 () is upregulated in mouse models of Juvenile X-linked Retinoschisis (XLRS), however no functional role for in disease progression has been identified. We performed electroretinogram (ERG) and standardized optical coherence tomography (OCT) in mice deficient in the Retinoschisin-1 () and and Caspase-11 genes (-KO ) to test the hypothesis that may play a role in disease evolution and or severity of disease. Currently, no studies have ventured to investigate the longer-term effects of on phenotypic severity and disease progression over time in XLRS, and specifically the effect on electroretinogram.

Article Synopsis
  • * The research utilized transcriptome data and animal models to investigate pyroptosis-related genes and potential drugs linked to UC by analyzing various microarray datasets and employing advanced analytical techniques like GSEA and WGCNA.
  • * A total of 20 pyroptosis-related differentially expressed genes (DEGs) were identified in UC, with 6 key candidate genes selected, leading to potential drug screening and experimental validation in animal studies.
