Ebola virus disease is one of the most serious and deadliest infectious diseases in the world with a sufficient number of recorded deaths. Since its discovery in 1976 until its last epidemic (14th), recorded and declared on April 23, 2022 by the National Minister of Public Health of the Democratic Republic of Congo and we have observed a succession of simultaneous epidemics of the disease to Ebola virus. This disease escapes the control of the health system of the Democratic Republic of the Congo, a health system which is already destabilized by the corona virus disease 2019 pandemic.

Ebola virus (EBOV), one member of the family Filoviridae, can causes hemorrhagic fever and other severe diseases in humans with a high mortality rate (25-90%). Until recently, there were no approved drugs and very limited treatment method for Ebola virus disease. In this study, we discovered a series of potent Ebola entry inhibitors with the (3S,4aS,8aS)-2-(3-amino-2-hydroxypropyl)decahydroisoquinoline-3-carboxamide scaffold from high-throughput screening in reported pseudotyped virus system.

The continuing emergence of viral pathogens and their rapid spread into heavily populated areas around the world underscore the urgency for development of highly effective vaccines to generate protective antiviral Ab responses. Many established and newly emerging viral pathogens, including HIV and Ebola viruses, are most prevalent in regions of the world in which infection remains endemic and vaccination at birth with bacille Calmette-Guérin (BCG) is widely used. We have investigated the potential for using CD4 T cells arising in response to BCG as a source of help for driving Ab responses against viral vaccines.
