Alkali treatment stabilizes fluctuations of urine AQP2 values measured by ELISA.

Background: Aquaporin-2 (AQP2) in urine is now measured in many water-balance disorders and regarded as a useful biomarker for diagnosis and prognosis. An enzyme-linked immunosorbent assay (ELISA) method has been developed for measurement of large numbers of clinical samples. However, fluctuations in the measured values were sometimes observed depending on storage conditions. Urine AQP2 is present in exosome membranes and we speculated that this structural organization causes the fluctuations.

Methods: Human urine samples from healthy subjects were measured by ELISA. Effects of maneuvers to disrupt the exosome membrane mechanically (freezing and thawing at different temperatures) and chemically (treating with alkali and detergents) prior to ELISA were examined.

Results: Urine samples stored at 4 or -80 °C did not show significant AQP2 values, whereas those stored at -25 °C for more that 2 weeks provided the values. Urine samples treated with 0.4 N NaOH and 0.5 % Triton X-305 showed the consistent and comparable values to those stored at -25 °C.

Conclusion: Pretreatment with alkali (0.4 N NaOH) to disrupt exosome membranes allowed consistent ELISA measurements of urinary AQP2. This simple method is applicable to ELISA of other membrane proteins included in exosomes.

Drying urine may open the opportunity to limit microbial and thermal degradation of the prohibited substances during transportation to the anti-doping laboratories without the need for refrigeration or freezing. Flow prediction in sound-based uroflowmetry.

Sound-based uroflowmetry (SU) offers a non-invasive alternative to traditional uroflowmetry (UF) for evaluating lower urinary tract dysfunctions, enabling home-based testing and reducing the need for clinic visits. This study compares SU and UF in estimating urine flow rate and voided volume in 50 male volunteers (aged 18-60), with UF results from a Minze uroflowmeter as the reference standard. Audio signals recorded during voiding were segmented and machine learning algorithms (gradient boosting, random forest, and support vector machine) estimated flow parameters from three devices: Ultramic384k, Mi A1 smartphone, and Oppo smartwatch.

β-cyclodextrin polymers as a new sorbent for solid-phase extraction of xenobiotics in Urine.

This study systematically assessed the performance of a newly developed solid-phase extraction (SPE) material, cellulose-supported aminated β-cyclodextrin polymer (amine-β-CDP@Cellulose), in determining 44 xenobiotics, encompassing endocrine-disrupting chemicals (EDCs), pharmaceuticals, and food additives in urine samples. The primary objective of the research was to synthesize a new sorbent, optimize the extraction protocol, and elucidate the underlying adsorption and desorption mechanisms. Following optimization, it was observed that amine-β-CDP@Cellulose achieved recoveries ranging from 80 % to 120 % for 28 of the 44 targeted xenobiotics, with only three compounds showing recoveries below 50 %.
