Picornaviruses are responsible for a range of human and animal diseases, but how their RNA genome is packaged remains poorly understood. A particularly poorly studied group within this family are those that lack the internal coat protein, VP4. Here we report the atomic structure of one such virus, Ljungan virus, the type member of the genus Parechovirus B, which has been linked to diabetes and myocarditis in humans. The 3.78-Å resolution cryo-electron microscopy structure shows remarkable features, including an extended VP1 C terminus, forming a major protuberance on the outer surface of the virus, and a basic motif at the N terminus of VP3, binding to which orders some 12% of the viral genome. This apparently charge-driven RNA attachment suggests that this branch of the picornaviruses uses a different mechanism of genome encapsidation, perhaps explored early in the evolution of picornaviruses.

Results: In this study, we developed a multiplex TaqMan probe-based real-time quantitative PCR (qPCR) assay for the rapid and simultaneous detection of kobuvirus, parechovirus B, rosavirus B and hunnivirus in murine rodent and shrew samples.

Background: To assess human parechovirus (HPeV) detection by real-time polymerase chain reaction in blood with or without cerebrospinal fluid samples in neonates and infants and compare the clinical and biological characteristics of HPeV patients with those of patients with or without enterovirus (EV) infection.
Methods: In all, 92 HPeV-infected patients (40 newborns and 52 infants) presenting with fever without source, sepsis-like disease or suspected meningitis were evaluated in 2 French hospitals from 2007 to 2018. The clinical and biological characteristics of HPeV patients were compared with those of 100 EV-infected patients and of 95 control patients with fever without source or sepsis-like disease.

Geckos are generally small, predominantly nocturnal reptiles, with several species commonly found close to human habitations. However, little is known about viral diversity in geckos. Using meta-transcriptomic sequencing we identified four novel RNA viruses - provisionally denoted Gecko astrovirus, Gecko parechovirus, Gecko reptillovirus, and Gecko hartmanivirus - in geckos sampled in October 2019 from Hainan Province, China.
