Purification and enzymatic characterization of a novel β-1,6-glucosidase from Aspergillus oryzae.

In this study, among the 10 genes that encode putative β-glucosidases in the glycoside hydrolase family 3 (GH3) with a signal peptide in the Aspergillus oryzae genome, we found a novel gene (AO090038000425) encoding β-1,6-glucosidase with a substrate specificity for gentiobiose. The transformant harboring AO090038000425, which we named bglH, was overexpressed under the control of the improved glaA gene promoter to form a small clear zone around the colony in a plate assay using 4-methylumbelliferyl β-d-glucopyranoside as the fluorogenic substrate for β-glucosidase. We purified BglH to homogeneity and enzymatically characterize this enzyme. The thermal and pH stabilities of BglH were higher than those of other previously studied A. oryzae β-glucosidases, and BglH was stable over a wide temperature range (4°C-60°C). BglH was inhibited by Hg(2+), Zn(2+), glucono-δ-lactone, glucose, dimethyl sulfoxide, and ethanol, but not by ethylenediaminetetraacetic acid. Interestingly, BglH preferentially hydrolyzed gentiobiose rather than other oligosaccharides and aryl β-glucosides, thereby demonstrating that this enzyme is a β-1,6-glucosidase. To the best of our knowledge, this is the first report of the purification and characterization of β-1,6-glucosidase from Aspergillus fungi or from other eukaryotes. This study suggests that it may be possible to find a more suitable β-glucosidase such as BglH for reducing the bitter taste of gentiobiose, and thus for controlling the sweetness of starch hydrolysates in the food industry via genome mining.

The protracted and immoderate utilization of chemical fertilizers has been detrimental to the composition of fungi in the soil and quality of crops. To ameliorate the adverse effects, a 6-year positioning experiment was undertaken to investigate the impact of substituting 0 % (CF), 25 % (M25), 50 % (M50), 75 % (M75), and 100 % (M100) of 225 kg ha chemical fertilizer nitrogen with manure nitrogen on both soil fungi and maize quality. This study showed that the expansion of Aspergillus heterocaryoticus, Xerochrysium dermatitidis, and Aspergillus penicillioides contributed to heightened levels of amylose and soluble sugars.

Starch-rich faba bean, yellow lentil, and yellow field pea flours were subjected to submerged fermentation using Aspergillus oryzae and Lactobacillus plantarum starter mono- or co-cultures, to increase protein contents of the flours. Fermentation mixes were supplemented with up to 35 g/L urea, ammonium sulfate and/or monoammonium phosphate as nitrogen sources. Protein contents of the flours increased 2-2.

Conventional light-driven antimicrobial strategies of zinc oxide (ZnO) are limited by inadequate illumination in dark environments. In this study, carboxylated cellulose nanocrystals (MCNC) mediated flower-like ZnO (C@Z) with self-promoted reactive oxygen species release under dark is fabricated. The adsorption of Zn ions on MCNC prompts the growth of ZnO along the (002) crystal plane, forming a flower-like hybrid with superior dispersibility and oxygen vacancies compared to MCNC-free ZnO, which exposes the (100) plane.

Airborne microorganisms in hospitals present significant health risks to both patients and employees. However, their pollution profiles and associated hazards in different hospital areas remained largely unknown during the extensive use of masks and disinfectants. This study investigated the characteristics of bioaerosols in an urban general hospital during the COVID-19 pandemic and found that airborne bacteria and fungi concentrations range from 87±35 to 1037±275 CFU/m and 21±15 to 561±132 CFU/m, respectively, with the outpatient clinic and internal medicine ward showing the highest levels.
