Synthesis of Spiroligomer-Containing Macrocycles.

We demonstrate the synthesis and characterization of the solution conformations of a collection of functionalized spiroligomer-based macrocycles. These macrocycles contain 14 independently controllable stereocenters and four independently controllable functional groups on a highly preorganized scaffold. These molecules are being developed to display complex, preorganized surfaces for binding proteins and to create enzyme-like active sites. In this work, we demonstrate the convergent synthetic approach to this new class of macrocycles and demonstrate that the conformational properties of these molecules can be changed by altering the configuration stereocenters within the backbone.

Structure-Based Design of "Head-to-Tail" Macrocyclic PROTACs.

Macrocyclization is a compelling strategy for conventional drug design for improving biological activity, target specificity, and metabolic stability, but it was rarely applied to the design of PROTACs possibly due to the mechanism and structural complexity. Herein, we report the rational design of the first series of "Head-to-Tail" macrocyclic PROTACs. The resulting molecule exhibited pronounced Brd4 protein degradation with low nM DC values while almost totally dismissing the "hook effect", which is a general character and common concern of a PROTAC, in multiple cancer cell lines.

Step Cyclization to Give Part Belt Oxygen-Functionalized Pillar[6,10]arenes.

Part belt oxygen-functionalized pillar[6,10]arenes are synthesized by step cyclization. An oxygen-substituted tetramer and hexamer have been prepared to construct the O3 pillar[6]arene via a hexamer cyclization reaction and the O6 pillar[10]arene through a [4 + 1+4 + 1] cyclization reaction. X-ray crystallographic studies reveal that both macrocycles have a large cavity and indicate that the part belt oxygen-functionalized pillar[6,10]arenes have both similar cyclic structures and certain differences in configuration compared to pillararene.

Objectives: Gadopiclenol is a q = 2 pyclen gadolinium-based contrast agent (GBCA) recently approved by the Food and Drug Administration, European Medicines Agency, and other European countries. The aim of this report is to demonstrate its stability in multiple stressed in vitro conditions and in vivo, in rat kidney, while maintaining its higher relaxivity compared with conventional GBCAs on the market.

Materials And Methods: Both gadopiclenol and its chemical precursor Pi828-Gd were characterized and compared with q = 1 gadolinium (Gd) complexes.

Background: Sheep scab, caused by the highly infectious mite, is considered to be endemic in Northern Ireland, although little investigation has been reported. A pilot project was undertaken to engage farmers, confirm cases with diagnostic methods and identify specific barriers to control, with the aim of informing future control programmes.

Methods: Through farmers self-reporting suspected outbreaks, on-farm risk assessments and clinical investigations were carried out by the farm's veterinary surgeon, who utilised light microscopy and serological testing to diagnose scab.
