Article Abstract

Human norovirus is a dominant cause of acute gastroenteritis around the world. Several norovirus disinfectants label citric acid as an active ingredient. In this study, we showed that norovirus virus-like particles (VLPs) treated with citrate buffer caused the particles to alter their morphology, including increased diameters associated with a new ring-like structure. We also found that epitopes on the protruding (P) domain on these particles were more readily accessible to antibodies after the citrate treatment. These results suggested that citrate had a direct effect on the norovirus particles. Using X-ray crystallography, we showed that the P domain bound citrate from lemon juice and a disinfectant containing citric acid. Importantly, citrate binds at the histo-blood group antigen binding pocket, which are attachment factors for norovirus infections. Taken together, these new findings suggested that it might be possible to treat/reduce norovirus infections with citrate, although further studies are needed.

The increased presence of resistant microorganisms in water promotes the need for supplementary measures to mitigate the water source's contamination. Traditional treatments are inefficient in wastewater management at removing some emerging contaminants. an invasive species, can be used in the treatment due to their resistance and biofiltration capacity, working as a pest management strategy.

The continually high disease burden of influenza and the relatively low effectiveness of current influenza vaccines call for enhanced vaccine strategies. We previously generated unique S-HA1 pseudovirus nanoparticles (PVNPs) displaying the receptor binding HA1 antigens of the H7N9 subtype as an influenza vaccine candidate and characterized their features in biochemistry, biophysics, structure, and immune response. In this follow up study, we created new S-HA1 PVNPs displaying the HA1 antigens of other common influenza viruses, including two H1N1 strains, one H3N2 strain, and an influenza B virus, respectively.

Magnetic ionic liquids (MILs) have proven effective as capture reagents for foodborne bacterial pathogens; however, there are currently no published studies regarding their use with foodborne, non-enveloped viruses. In this study, a protocol was evaluated for capture and recovery of bacteriophage MS2, a human norovirus surrogate, and purified viral genomic single stranded RNA (ssRNA) from an aqueous suspension using MILs. Transition metal-based MILs showed similar capture and recovery efficiency for both targets.

Human norovirus disturbs intestinal motility and transit time through its capsid proteins.

PLoS Pathog

Article Synopsis
  • Human norovirus (HuNoV) causes over 700 million cases of gastroenteritis yearly, leading to symptoms like vomiting and diarrhea, but the mechanisms of infection are not well understood due to the absence of suitable animal models.
  • Researchers utilized a zebrafish larvae model to investigate how HuNoV affects intestinal motility and whether a specific viral protein might act as an enterotoxin.
  • The study found that HuNoV GII.4 infection increased intestinal contraction frequency and delayed food transit time in the larvae, indicating potential effects on bowel movements, with viral proteins VP1 and VP2 playing significant roles in these symptoms.

Modular Display of Circumsporozoite Surface Protein and Merozoite Surface Protein-1 on Norovirus-like Particles.

Bioconjug Chem

December 2024

Laboratory of Biotechnology, Research Institute of Green Science and Technology, Shizuoka University, 836 Ohya Suruga-ku, Shizuoka 422-8529, Japan.

Recently, virus-like particles have been regarded as a promising platform for displaying foreign peptides or proteins on their surface. In this study, a dual-protein-displaying platform based on the norovirus-like particle (NoV-LP) was developed using SpyTag (SpT)/SpyCatcher (SpC) protein bioconjugation. A short 14-amino-acid SpT peptide was added to the C-terminus of VP1, with a rigid "EAAAK" spacer in between.
