HisA is a (βα)8 barrel enzyme that catalyzes the Amadori rearrangement of N'-[(5'-phosphoribosyl)formimino]-5-aminoimidazole-4-carboxamide ribonucleotide (ProFAR) to N'-((5'-phosphoribulosyl) formimino)-5-aminoimidazole-4-carboxamide-ribonucleotide (PRFAR) in the histidine biosynthesis pathway, and it is a paradigm for the study of enzyme evolution. Still, its exact catalytic mechanism has remained unclear. Here, we present crystal structures of wild type Salmonella enterica HisA (SeHisA) in its apo-state and of mutants D7N and D7N/D176A in complex with two different conformations of the labile substrate ProFAR, which was structurally visualized for the first time. Site-directed mutagenesis and kinetics demonstrated that Asp-7 acts as the catalytic base, and Asp-176 acts as the catalytic acid. The SeHisA structures with ProFAR display two different states of the long loops on the catalytic face of the structure and demonstrate that initial binding of ProFAR to the active site is independent of loop interactions. When the long loops enclose the substrate, ProFAR adopts an extended conformation where its non-reacting half is in a product-like conformation. This change is associated with shifts in a hydrogen bond network including His-47, Asp-129, Thr-171, and Ser-202, all shown to be functionally important. The closed conformation structure is highly similar to the bifunctional HisA homologue PriA in complex with PRFAR, thus proving that structure and mechanism are conserved between HisA and PriA. This study clarifies the mechanistic cycle of HisA and provides a striking example of how an enzyme and its substrate can undergo coordinated conformational changes before catalysis.

Aim: Despite dysfunctional vasoactive intestinal polypeptide-positive interneurons (VIP-INs) being linked to the emergence of neurodevelopmental disorders, the temporal profile of VIP-IN functional maturation and cortical network integration remains unclear.
Methods: Postnatal VIP-IN development was traced with patch clamp experiments in the somatosensory cortex of Vip-IRES-cre x tdTomato mice. Age groups were chosen during barrel field formation, before and after activation of main sensory inputs, and in adult animals (postnatal days (P) P3-4, P8-10, P14-16, and P30-36).
Prairie voles (Microtus ochrogaster) are one of the few mammalian species that are monogamous and engage in the biparental rearing of their offspring. Biparental care impacts the quantity and quality of care the offspring receives. The increased attention by the father may translate to heightened tactile contact the offspring receives through licking and grooming.

Since the release of the first 3D-printed firearm, "The Liberator," the occurrence of 3D-printed firearms in criminal activities has increased, highlighting the need for forensic research on these weapons. This study presents a technical examination of a 3D-printed firearm received by the National Institute of Criminalistics and Criminology (NICC), focusing on its design, ballistic performance, and its potential for microscopic comparative analysis. The firearm, resembling a 3D-printed pistol Yeet22, is primarily constructed from polymer parts, with the exception of the firing pin, barrel, and various springs and screws.

The use of nanozymes for electrochemical detection in the food industry is an intriguing area of research. In this study, we synthesized a laccase mimicking the MnO@CeO nanozyme using a simple hydrothermal method, which was characterized by modern analytical methods, such as transmission electron microscope (TEM), X-ray diffraction (XRD), and energy dispersive X-ray spectroscopy (EDX), etc. We found that the addition of MnO significantly increased the laccase-like activity by 300% compared to CeO nanorods.

The aim of the study was to determine the relationship between slaughter weight (SW) with body components and liner body measurements and investigate the coefficient of correlation between slaughter weight with body component and liner body measurements to select the best regression equation. Data on liner body measurements (height at wither and at hips, heart girth, body length, height and width of hump, height at fall and hind legs, body sheath height, height at hooks, barrel circumference, width of face, length of face and tail circumference) and slaughter weight of body components (Hot Carcass Weight (HCW), Empty Body Weight (ESW), Internal Offal (IO) and External Offal (EO)) were collected from 62 Hararghe cattle at Haramaya University abattoir. ESW was calculated as SW with less gut contents.
