Colistin has re-emerged as an essential antibiotic for the treatment of carbapenem-resistant Gram-negative infections. Unfortunately, its utility is limited by high rates of nephrotoxicity, even at potentially therapeutic concentrations, and an overall lack of understanding on how to optimally administer the agent. In this review, recent advancements in the understanding of the safety and efficacy of colistin are discussed and strategies and suggestions on how to balance the two are described.

Background: Despite advancements in the therapeutic armamentarium for Crohn's disease (CD), biologic and small molecule monotherapies are associated with sub-optimal response and remission rates. Utilizing dual biologic therapy (DBT) holds the potential to increase efficacy in the treatment of refractory or partially responsive CD. Evidence pertaining to this strategy remains limited.

Magnetorheological (MR) fluid (MRF) dampers, serving as fail-safe semi-active devices, exhibit nonlinear hysteresis characteristics, emphasizing the necessity for accurate modeling to formulate effective control strategies in smart systems. This paper introduces a novel stop operator-based Prandtl-Ishlinskii (PI) model, featuring a reduced parameter set (seven), designed to estimate the nonlinear hysteresis properties of a large-scale bypass MRF damper with variable stiffness capabilities under varying applied current. With only seven parameters, the model realizes current, displacement, and rate dependencies.

Introduction Lumbar pyogenic spondylodiscitis is a challenging and rare spinal infection with high morbidity, particularly in patients with comorbidities. While the extreme lateral interbody fusion (XLIF) technique is established in treating degenerative spinal conditions, its efficacy in managing spondylodiscitis is less well-studied. This study aims to evaluate the clinical and radiographic outcomes of the XLIF approach combined with posterior instrumentation in patients with lumbar spondylodiscitis.

Background: Lower respiratory tract infections (LRTIs) pose a significant threat to global health, causing more than 2 million deaths worldwide. This menace is intensified by the alarming increase in drug resistance, which limits the availability of effective antibiotics for bacterial respiratory infections. Consequently, there is an urgent demand for alternative therapeutic options.

Emerging evidence demonstrates that inducing ferroptosis, a nonapoptotic programmed cell death mode, holds significant potential for tumor treatment. However, current ferroptosis strategies utilizing exogenous Fenton-type heavy metal species or introducing glutathione (GSH)/glutathione peroxidase 4 (GPX4) suppressants are hampered by latent adverse effects toward organisms, while utilizing endogenous iron may cause undesirable tumor angiogenesis through specific signaling pathways. Here, a ferric ion (Fe)-responsive and DNAzyme-delivered coordination nanosystem (ZDD) is developed to achieve a novel scheme of synergistic tumor-specific ferroptosis and gene therapy, which modulates and harnesses the endogenous iron in tumors for inducing ferroptosis while intercepting tumor angiogenesis to enhance therapeutic efficacy.
