Osteopontin is robustly upregulated following myocardial infarction (MI), which suggests that it has an important role in post-MI remodeling of the left ventricle (LV). Osteopontin deletion results in increased LV dilation and worsened cardiac function. Thus, osteopontin exerts protective effects post-MI, but the mechanisms have yet to be defined. Matrix metalloproteinases (MMPs) regulate LV remodeling post-MI, and osteopontin is a known substrate for MMP-2, -3, -7, and -9, although the cleavage sites have not been mapped. Osteopontin-derived peptides can exert distinct biological functions that may depend on their cleavage sites. We mapped the MMP-9 cleavage sites via LC-MS/MS analysis using label-free and N-terminal labeling methods, and compared them with those of MMP-2, -3, and -7. Each MMP yielded a unique cleavage profile with few overlapping cleavage sites. Using synthetic peptides, we validated 3 sites for MMP-9 cleavage at amino acid positions 151-152, 193-194, and 195-196. Four peptides were synthesized based on the upstream- and downstream-generated fragments and were tested for biological activity in isolated cardiac fibroblasts. Two peptides increased cardiac fibroblast migration rates post-wounding (p < 0.05 compared with the negative control). Our study highlights the importance of osteopontin processing, and confirms that different cleavage sites generate osteopontin peptides with distinct biological functions.

Localized prostate cancer can be cured by radiation or surgery, but advanced prostate cancer continues to be a clinical challenge. Altered alternative polyadenylation occurs in numerous cancers and can downregulate tumor-suppressor genes and upregulate oncogenes. We found that the cleavage and polyadenylation specificity factor (CPSF) complex factor CPSF1 is upregulated in patients with advanced prostate cancer, with high CPSF1 expression correlating with worse progression-free survival.

In the CO reduction reactions (CORR), the product selectivity is strongly dependent on the binding energy differences of the key intermediates. Herein, we systematically evaluated the CORR reaction pathways on single transition metal atom doped catalysts TMCu/CuO by density functional theory (DFT) methods and found that *CO is more likely to undergo C-O bond cleavage rather than be hydrogenated on TMCu/CuO (TM = Sc, Ti, V, Cr, Mn, Fe, Co), which facilitates C production with a low-energy pathway of OC-C coupling, while it prefers to be hydrogenated to form CHO on TMCu/CuO (TM = Ni, Cu). The defects of Cu in TMCu/CuO were confirmed to enhance the production of ethanol.

Building anion-derived solid electrolyte interphase (SEI) with enriched LiF is considered the most promising strategy to address inferior safety features and poor cyclability of lithium-metal batteries (LMBs). Herein, we discover that, instead of direct electron transfer from surface polar groups to bis(trifluoromethanesulfonyl)imide (TFSI) for inducing a LiF-rich SEI, the dipole-induced fluorinated-anion decomposition reaction begins with the adsorption of Li ions and is highly dependent on their mobility on the polar surface. To demonstrate this, a single-layer graphdiyne on MXene (sGDY@MXene) heterostructure has been successfully fabricated and integrated into polypropylene separators.
