Nitroxide malonate methanofullerene as biomimetic model of interaction of nitroxide species with antioxidants.

A.E. Arbusov Institute of Organic and Physical Chemistry, Kazan Scientific Center, Russian Academy of Sciences, Arbuzov str. 8, 420088 Kazan, Russian Federation.

Bis-nitroxide malonate methanofullerene (NO)2-MF was studied as a biomimetic model of reduction-oxidation activity with natural compounds-cytochrome c (cyt c), dihydroquercetin (DHQ), ascorbic acid (AA) and synthetic drug-1-(β-oxyethyl)-4,6-dimethyl-1,2-dihydro-2-oxopyrimidine (xymedon(®)). (NO)2-MF may be used as the component of Langmuir monolayers on an aqueous subphase and as the adsorbate on silica gel. The activity of (NO)2-MF in the reaction with cyt c was compared with the effect of nitroxide species such as gaseous nitric oxide, 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO) by using UV-vis and EPR-spectra.

Using the model of rat sciatic nerve transection and crush injury we studied influence of pyrimidine derivative xymedon on efficacy of regeneration of myelinated axons, number and phenotype of surviving sensory neurons (expressing GAP-43 and Bcl-2) and Schwann cells (S100, GAP-43, PCNA) on the 7th, 15th, 30th, 60th and 90th day after nerve injury. We found out that xymedon counteracts posttraumatic death of sensory neurons, stimulates regeneration of myelinated fibres and proliferation of Schwann cells.

An investigation of the antioxidant activity of blood plasma allowed a conclusion to be made of an alteration of the free radical processes in patients with chronic osteomyelitis. Experimental data on the influence of the antioxidants Xymedon and Methyluracil on the main free radicals, lipid peroxidation and chemiluminescence of peritoneal macrophages of rats allowed the substantiation of using the medicines in the complex treatment of patients in the pre- and postoperative periods. As a result, in 25 patients Xymedon improved the blood indices, increased antioxidant activity of plasma and reduced the amount of recurrences of the disease as compared with the patients taking Methyluracil.
