The first example of an ammonium derivative that causes a specific modification of the active site of type I dehydroquinase (DHQ1), a dehydratase enzyme that is a promising target for antivirulence drug discovery, is described. The resolution at 1.35 Å of the crystal structure of DHQ1 from Salmonella typhi chemically modified by this ammonium derivative revealed that the ligand is covalently attached to the essential Lys170 through the formation of an amine. The detection by mass spectroscopy of the reaction intermediates, in conjunction with the results of molecular dynamics simulations, allowed us to explain the inhibition mechanism and the experimentally observed differences between S. typhi and Staphylococcus aureus enzymes. The results presented here reveal that the replacement of Phe225 in St-DHQ1 by Tyr214 in Sa-DHQ1 and its hydrogen bonding interaction with the conserved water molecule observed in several crystal structures protects the amino adduct against further dehydration/aromatization reactions. In contrast, for the St-DHQ1 enzyme, the carboxylate group of Asp114, with the assistance of this water molecule, would trigger the formation of a Schiff base that can undergo further dehydration reactions until full aromatization of the cyclohexane ring is achieved. Moreover, in vitro antivirulence studies showed that the reported compound is able to reduce the ability of Salmonella Enteritidis to kill A459 respiratory cells. These studies have identified a good scaffold for the design of irreversible inhibitors that can be used as drugs and has opened up new opportunities for the development of novel antivirulence agents by targeting the DHQ1 enzyme.

This study focuses on the synthesis, characterization, and evaluation of the photocatalytic efficiency of bismuth-based metal-organic frameworks (Bi-MOFs) and their derivatives, specifically Ag/Bi-MOF and NH /Ag/Bi-MOF, in the degradation of tetracycline (TC) and sulfamethoxazole (SMX) under visible light irradiation. Bi-MOFs are promising photocatalysts due to their large surface area, tunable porosity, and unique electronic properties that are favorable for visible light absorption. In this study, Bi-MOFs were synthesized using a solvothermal method, with the incorporation of silver (Ag) and ammonium (NH ) ions to enhance their photocatalytic performance.

A human oral phase followed by a dynamic gastrointestinal digestion and colonic fermentation (simgi®) has been applied to wholemeal rye bread (WRB) and PS-enriched WRB (PS-WRB). The aim of this study was to evaluate the impact of these solid and high-fiber food matrices on the metabolism of PS, modulation of the microbiota and production of short-chain fatty acids (SCFA) and ammonium ion after a simulated chronic intake (5 days). In both breads, campesterol, campestanol, stigmasterol, β-sitosterol, sitostanol, Δ5-avenasterol, Δ5,24-stigmastadienol, Δ7-stigmastenol, and Δ7-avenasterol were identified, of which only β-sitosterol was metabolized to sitostenone after PS-WRB treatment.

Greywater, originating from kitchen sinks and toilets, constitutes 75-80 % of the domestic wastewater produced in homes and can be reclaimed for non-potable uses. This study synthesized novel sludge-derived aluminosilicates and alginate-polyethyleneimine (PEI) biochar composites. The aluminosilicates offer a sustainable approach to sludge management, while alginate-polyethyleneimine presents a green biochar modification approach.

These herbicides give rise to a number of key metabolites, with 3-methyl phosphinicopropionic acid (MPPA), -acetyl glufosinate (-acetyl GLUF), aminomethyl phosphonic acid (AMPA), -acetyl aminomethyl phosphonic acid (-acetyl AMPA), -acetyl glyphosate (-acetyl GLY), -methyl glyphosate (-methyl GLY) as the major metabolites obtained from GLUF and GLY. Extensive use of these herbicides may lead to their increased presence in the environment, especially aquatic ecosystems.

Thromboxane A (TXA), a prothrombotic factor that induces platelet aggregation and thrombosis, acts as a vasoconstrictor by activating TXA receptors (TP receptors). TXA is extremely unstable and metabolizes into three major metabolites: 2,3-dinor thromboxane B (2,3-dinor-TXB), 11-dehydro TXB(11-dh-TXB), and 11-dehydro-2,3-dinor TXB(11-dh-2,3-dinor-TXB). 8-Iso-prostaglandin F(8-iso-PGF), a prostaglandin-like compound widely considered the best biomarker of oxidative stress, can also activate TP receptors.
