Objective: To analyze the influence of miRNA-184 on epithelial-mesenchymal transition (EMT) of human lens epithelial cells (HLEC) induced by TGF-beta2 in vitro.

Methods: Experimental study. Recombinant plasmid of pL/IRES/GFP-miR-184 was constructed and used to produce the lentivirus. The lentivirus was used to transduce the HLEC which was in the process of EMT induced by transforming growth factor-β2 (TGF-β2). The real-time fluorescence quantitative PCR (QRT-PCR) was used to analyze E-cadherin (CDH1), α-smooth muscle actin (α-SMA), vimentin (VIM) expression at RNA levels during interval of 0 h, 6 h and 24 h after transduction, in comparison with that of control group. Statistical analysis method was single factor variance analysis.

Results: The expression level of epithelial marker gene CDH1 in the miRNA-184 transduced group maintains relatively stable during 24h interval, while it goes down in the control group. The expression level of mesenchymal cell marker gene VIM, α-SMA in the miRNA-184 transduced group maintain relatively stable, while it goes up in the control group. The results of statistical analysis showed a statistically significant difference between miRNA-184 transduced group and control group (P<0.05).

Conclusion: miRNA-184 lentivirus-mediated HLEC can inhibit the occurrence of EMT.
