Orthogonal bond-breaking and forming of dynamic covalent disulfide and imine bonds in aqueous solution is demonstrated. Through judicious choice of reaction partners and conditions, it is possible to cleave and reform selectively these bonds in the presence of each other in the absence of unwanted competing processes.

Microbial mats are stratified communities often dominated by unicellular and filamentous phototrophs within an exopolymer matrix. It is challenging to quantify the dynamic responses of community members in situ as they experience steep gradients and rapid fluctuations of light. To address this, we developed a binary consortium using two representative isolates from hot spring mats: the unicellular oxygenic phototrophic cyanobacterium OS-B' (Syn OS-B') and the filamentous anoxygenic phototroph MS-CIW-1 (Chfl MS-1).

Yet, several inherent limitations restrict applying the approaches to genes not suitable for RNA velocity inference due to complex transcriptional dynamics, low expression, or lacking splicing dynamics, or data of non-transcriptomic modality. Here, we present GraphVelo, a graph-based machine learning procedure that uses as input the RNA velocities inferred from existing methods and infers velocity vectors lying in the tangent space of the low-dimensional manifold formed by the single cell data.
